Pan-Cancer landscape of protein activities identifies drivers of signalling dysregulation and patient survival

Summary: Genetic alterations in cancer cells lead to dysregulation of kinase and transcription factor activities, contributing to oncogenic transformation. We analyzed genomics and (phospho)proteomics data from a large cohort of tumors and cell lines to estimate activity changes in hundreds of kinases and transcription factors. We found co-regulation of kinase and TF activities, revealing known regulatory relationships and dissecting genetic drivers of signaling changes in cancer. Our study also identified differentially regulated activities in cancer subtypes and their association with patient survival, providing insights into the dysregulation of protein activities and its impact on disease severity in cancer.

Hallmarks of Cancer: New Dimensions

This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Discovery of a selective c-MET inhibitor with a novel binding mode

Summary: In this study, a highly selective c-MET inhibitor was discovered through screening a DNA-encoded chemical library, and it was found to bind to the kinase in an unprecedented manner as shown by X-ray crystallography. This novel mode of inhibiting c-MET with a small molecule may offer a potential approach to target drug-resistant forms of the kinase while avoiding potential toxicity issues associated with broad kinome inhibition.

Summary: This study developed sulfuretin analogs that have the potential to inhibit the immunosuppressive PGE(2) and directly inhibit the growth of Leishmania donovani. Among the library of analogs, compounds 1i and 1q showed the most promising activity. Cytotoxicity evaluation demonstrated that compound 1i specifically targets the parasite without affecting host cells. In silico simulation provided insights into the binding between these analogs and Leishmania donovani fumarate reductase. These findings suggest that compounds 1i and 1q could be further developed as multifunctional therapeutic agents against visceral leishmaniasis.
