Exosomal microRNAs array sensor with a bioconjugate composed of p53 protein and hydrazine for the specific lung cancer detection

Summary: This study demonstrates a novel strategy for the early diagnosis of lung cancer by detecting microRNAs encapsulated in exosomes using immunomagnetic isolation. The researchers selectively extracted exo-miRNAs from patient serum by capturing miRNA from lysed exosomes with specially designed capture probe modified magnetic beads. The sensor probe formed through T4 DNA polymerase-mediated in situ formation of chimeric 5'-miRNA-DNA-3' (Target) and p53 protein recognition showed excellent analytical performance, with a dynamic range between 100 aM and 10 pM and a detection limit of 92 (+/- 0.1) aM. Furthermore, a multiplexed array sensor was prepared to detect four exo-miRNAs at femtomolar levels in cell culture media and lung cancer patient serum samples.

Autoantibody panel on small extracellular vesicles for the early detection of lung cancer

Summary: This study evaluated the potential use of autoantibodies on sEVs as a biomarker for early detection of lung cancer. The results showed that the autoantibody panel on sEVs was more accurate in distinguishing between benign lung disease and lung cancer compared to serum. Further analysis demonstrated that the autoantibodies could bind to antigens on sEVs, potentially contributing to immune escape of lung cancer.

This sensor offers a conjugation-free, non-enzymatic Exo-miR detection in blood and accurately distinguishes breast cancer patients from normal individuals, showing promise for early tumor diagnosis.

Peptide Nucleic Acid-Functionalized Nanochannel Biosensor for the Highly Sensitive Detection of Tumor Exosomal MicroRNA

Summary: A PNA-functionalized nanochannel biosensor was proposed for the ultrasensitive and specific detection of tumor exosomal miRNAs, achieving a detection limit as low as 75 aM. The biosensor could effectively distinguish pancreatic cancer tumor-derived exosomes from the normal control group, and the results were consistent with qRT-PCR method. Moreover, the biosensor showed potential for early cancer diagnosis by detecting exosomal miRNA-10b in clinical plasma samples with higher content in cancer patients compared to healthy individuals.

Summary: Metal-loaded hollow carbon nanostructures (MHC nanoreactors) exhibit competitive potentials in heterogeneous catalysis due to their tailorable microenvironment effects, with the void-confinement effect being one of the most fundamental functions. The void-confinement effect of the selected MHC nanoreactors is essentially driven by an integrating action of electronic metal-support interaction, reactant enrichment and diffusion, showcasing the unique properties of hollow nanoreactors in electronic and structural aspects. This work offers insights into the catalytic behavior of MHC nanoreactors and potential strategies for microenvironment engineering of hollow nanostructures.
