Reversible RNA phosphorylation stabilizes tRNA for cellular thermotolerance

Summary: Post-transcriptional modifications play critical roles in tRNA stability and function. A 2'-phosphouridine modification at position 47 of tRNAs in thermophilic archaea was identified, which confers thermal stability and nuclease resistance to tRNAs. The modification is mediated by the enzyme ArkI and dephosphorylated by the enzyme KptA, and it fine-tunes the structural rigidity of tRNAs under extreme environmental conditions.

Epitranscriptomic Reprogramming Is Required to Prevent Stress and Damage from Acetaminophen

Summary: In this study, we investigate how acetaminophen induces epitranscriptomic reprogramming and the role of the writer enzyme Alkylation Repair Homolog 8 (Alkbh8) in gene regulation during this response. We find that APAP promotes large-scale reprogramming of various RNA marks, and deficiency in Alkbh8 leads to widespread dysregulation of the epitranscriptome. Our findings highlight the importance of RNA modifications and translational responses to pharmaceuticals.

Arsenite toxicity is regulated by queuine availability and oxidation-induced reprogramming of the human tRNA epitranscriptome

Summary: Cells respond to environmental stress by regulating gene expression at the level of both transcription and translation. The dynamic regulation of transfer RNA (tRNA) wobble modifications leads to selective translation of stress response proteins. This study reveals that the response of human hepatocellular carcinoma cells to arsenite exposure is regulated by the availability of queuine, a precursor to tRNA wobble modification. Limiting queuine leads to cell death, altered translation, increased reactive oxygen species levels, and mitochondrial dysfunction.
