Structural elucidation and relative quantification of sodium- and potassium-cationized phosphatidylcholine regioisomers directly from tissue using electron induced dissociation

Separation of Isobaric Lipids in Imaging Mass Spectrometry Using Gas-Phase Charge Inversion Ion/Ion Reactions

Summary: The diverse array of chemical compounds in tissue samples can result in many isobaric compounds in imaging mass spectrometry experiments. High-resolution accurate mass measurements can help resolve these compounds, but may not always be feasible. Alternatively, tandem mass spectrometry can be used to improve molecular specificity. Gas-phase ion/ion reactions can selectively react with desired analytes and differentiate isobaric compounds. This method has been used to differentiate isobaric lipids in the imaging mass spectrometry analysis of rat brain tissue.

Summary: The application of matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) to investigate the changes of lipids in biological tissues has yielded significant results. However, the presence of structural isomers with varying C=C double bond (DB) locations calls for the development of isomer-resolved MSI. In this study, a new light-driven on-tissue [2 + 2] cycloaddition reaction coupled with MALDI-MS/MS imaging was introduced to identify lipid DB position isomers and their spatial signatures in biological tissues.

Unsaturated lipid isomeric imaging based on the Paterno-Buchi reaction in the solid phase in ambient conditions

Summary: The development of unsaturated lipid isomeric imaging based on the Patern`o-Buchi (PB) reaction has significantly improved in recent years. A method of PB reaction in the solid phase in ambient conditions was developed in this paper, combined with air-flow-assisted desorption electrospray ionisation mass spectrometry (AFADESI-MS) to achieve in situ imaging of lipids at an isomeric level. This method simplifies reaction conditions in the imaging process and may be useful for future studies on lipid isomers in tissues.

Summary: Prostate cancer diagnosis relies on biopsy and histopathology, which can vary between pathologists, leading to investigation of biomolecular markers. This study used isomer-resolved lipidomics and imaging mass spectrometry to explore lipid metabolic signatures in prostate cancer tissue sections, finding correlations between lipid isomers and pre-malignant lesions and inflammation. The results suggest a potential role for isomer-resolved mass spectrometry imaging in assisting pathologists with prostate cancer diagnoses and patient stratification.
