Artificial biosynthetic pathways utilizing enzymes have the ability to carry out multiple synthetic steps simultaneously, enabling the discovery and production of complex, non-natural molecules. In this study, an enzymatic cascade was discovered and constructed for the synthesis of MK-1454, a potential immuno-oncology therapeutic. By engineering the enzymes involved, multiple steps of the synthesis were successfully carried out in a one-pot reaction.

Intercellular transfer of activated STING triggered by RAB22A-mediated non-canonical autophagy promotes antitumor immunity

Summary: Activated STING can be transferred between cells to promote antitumor immunity, a process triggered by non-canonical autophagy mediated by RAB22A. RAB22A induces the fusion of non-canonical autophagosomes with early endosomes, forming a new organelle called Rafeesome, and suppresses fusion with lysosomes. The inner vesicle of non-canonical autophagosome, containing activated STING, is secreted as R-EV, which induces the release of IFN beta from recipient cells and promotes antitumor immunity.

Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

TCR-engineered T cells targeting E7 for patients with metastatic HPV-associated epithelial cancers
