Exogenous hydrogen sulfide (H2S) exerts therapeutic potential in triple-negative breast cancer by affecting cell cycle and DNA replication pathway

Xiaoyue Cui et al.

Summary: Exogenous hydrogen sulfide (H2S) has been found to inhibit the proliferation, migration, and invasion of triple negative breast cancer (TNBC) cells, and decrease the malignancy of TNBC in a mouse model. Further analysis revealed that exogenous H2S exerts its anti-TNBC effects by delaying DNA replication.

Metformin and Cancer: Solutions to a Real-World Evidence Failure

Oriana Hoi Yun Yu et al.

Summary: The repurposing of metformin as a cancer prevention and treatment agent generated extensive research. Observational studies reported a reduction in cancer incidence, but these findings were later found to be biased. Randomized trials also did not find reductions in cancer incidence. Therefore, rigorous approaches are needed for observational studies to evaluate drug repurposing.

Erucin, an H2S-Releasing Isothiocyanate, Exerts Anticancer Effects in Human Triple-Negative Breast Cancer Cells Triggering Autophagy-Dependent Apoptotic Cell Death

Ivana Bello et al.

Summary: Breast cancer is the most common cancer in women, and triple-negative breast cancer (TNBC) is the most severe subtype with the highest mortality rate. This study explored the potential anti-cancer effects of erucin, a compound found in arugula, on TNBC cells. The results showed that erucin effectively inhibited TNBC cell proliferation, induced apoptosis and autophagy, prevented ROS generation, and inhibited cell migration, invasion, and colony formation. Therefore, erucin consumption could be a promising strategy for TNBC intervention.

Hydrogen Sulfide (H2S): As a Potent Modulator and Therapeutic Prodrug in Cancer

Pawan Faris et al.

Summary: Hydrogen sulfide (H2S) is an endogenous gaseous molecule studied for its toxicity, but recent research has shown its relevance as a signaling molecule with potential implications in clinical disorders and cancer. This article provides an overview of H2S's properties, enzymatic pathways, and role as a signaling molecule. It also discusses the potential of H2S-releasing agents as therapeutic tools, the paradoxical role of H2S in tumor biology, and the use of H2S levels as cancer biomarkers. Additionally, it describes the preclinical applications of H2S-releasing compounds in assessing their anti-cancer impact.

Hydrogen sulfide: Recent development of its dual donors and hybrid drugs

Shi Xu et al.

Summary: Hydrogen sulfide (H2S) is an important gaseous signaling molecule that regulates cellular redox homeostasis. The development of H2S-releasing compounds, including dual donors and hybrid drugs, has emerged to harness the therapeutic potential of H2S in various diseases. These compounds aim to offset negative side effects and achieve synergistic effects, providing more clinically effective treatments. This review focuses on recent advances in the design and effects of H2S-based dual donors and hybrid drugs.

Prednisone-hydrogen sulfide releasing hybrid shows improved therapeutic profile in asthma

Ida Cerqua et al.

Summary: This study investigated the effectiveness of a novel corticosteroid derivative chemically linked to an H2S donor in managing asthma features. The results showed that the combination of prednisone and the H2S donor had increased effects in modulating inflammatory mediators and controlling airway hyperreactivity compared to the individual compounds. Moreover, the combination therapy also demonstrated efficacy in controlling airway remodeling, promoting epithelial repair, and inhibiting mucin production.

Rosa Sparaco et al.

Summary: This study designed and synthesized new molecular hybrids that combine antiglaucoma drugs with H2S donors, aiming to provide better treatment effects by releasing hydrogen sulfide. Experimental results showed that certain compounds can significantly and persistently produce hydrogen sulfide in both aqueous solution and intracellular environment. These findings lay the foundation for further investigation on the efficacy of the newly synthesized compounds.
