Transcriptome analysis reveals dysregulation of inflammatory and neuronal function in dorsal root ganglion of paclitaxel-induced peripheral neuropathy rats

Summary: Chemotherapy-induced peripheral neuropathy (CIPN) is a common side-effect of anti-cancer therapy. The exact pathogenesis of CIPN is still unclear, and there are no effective analgesics for prevention and treatment. In this study, a paclitaxel-induced peripheral neuropathy (PIPN) model was used to investigate the transcriptomic level of DRG neurons in rats with PIPN. RNAseq analysis identified differentially expressed genes (DEGs) related to neuroactive ligand-receptor interaction, cytokine-cytokine receptor interaction, defense response, and inflammatory response. The study also found shared characteristics of PIPN with inflammatory pain and neuropathic pain.

MicroRNA-133b-3p Targets Purinergic P2X4 Receptor to Regulate Central Poststroke Pain in Rats

Summary: In this study, the authors found that the expression of Purinergic P2X4 receptor (P2X4R) was associated with the downregulation of microRNA-133b-3p (miR-133b-3p) in patients with central poststroke pain (CPSP). The downregulation of miR-133b-3p was negatively correlated with the severity of allodynia. Genetic knockdown of P2X4R and genetic overexpression of miR-133b-3p alleviated allodynia in CPSP rats. Additionally, treatment with gabapentin restored the expression of miR-133b-3p and blocked allodynia in CPSP rats.

Plasticity of neuronal excitability and synaptic balance in the anterior nucleus of paraventricular thalamus after nerve injury

Summary: The excitability and synaptic inputs of aPVT neurons are affected in neuropathic pain, as shown by the increase in neuronal excitability and changes in the excitatory/inhibitory synaptic inputs ratio after spinal nerve ligation (SNL). Additionally, SNL alters the physiological properties of aPVT neurons, such as membrane input resistance and action potential characteristics. These findings suggest that the plasticity of aPVT neurons plays a crucial role in the development of pain sensitization after nerve injury.
