Ribosome biogenesis factors-from names to functions

Summary: The assembly of ribosomal subunits is a highly coordinated process involving many accessory factors. Most of these factors have been identified through genetic screening and proteomic approaches in yeast, and their requirement is conserved in evolution. However, the function and mechanism of action of many factors remained unknown until recently, when structural, biochemical, and cellular studies provided a detailed understanding of their role in ribosome maturation.

Principles of human pre-60S biogenesis

Assembly landscape for the bacterial large ribosomal subunit

Emergence of the primordial pre-60S from the 90S pre-ribosome

Summary: The synthesis of ribosomes in the nucleolus involves the formation of pre-ribosomes, from which the 40S and 60S pathways diverge. However, it is unknown how the earliest pre-60S subunit continues to develop after this separation. This study reveals a large-subunit intermediate, connected to the 90S pre-ribosome, that contains specific proteins and RNAs. The inability to disconnect from the 90S in a mutant allows for the visualization of a 70nm 90S-pre-60S particle. This research provides insights into the assembly pathway of the still-connected nascent 40S and 60S subunits.

Wide mutational analysis to ascertain the functional roles of eL33 in ribosome biogenesis and translation initiation

Summary: An extensive mutational analysis of the RPL33A gene encoding yeast ribosomal protein L33A (eL33) identified several novel rpl33a mutants with different translational phenotypes. These mutants exhibit defects in pre-rRNA processing and mature rRNA production, leading to reductions in ribosomal subunit abundance and altered polysome profiles. Some mutants also show constitutive derepression of GCN4 translation and slow growth phenotypes. Interestingly, certain mutants exhibit an increase in the UUG/AUG translation initiation ratio, which can be suppressed by eIF1 overexpression.
