Selective Photo-Assisted Eradication of Triple-Negative Breast Cancer Cells through Aptamer Decoration of Doped Conjugated Polymer Nanoparticles

Luis Exequiel Ibarra et al.

Summary: Photodynamic therapy (PDT) may be a promising alternative for the treatment of breast cancer, especially for aggressive types like triple-negative breast cancer (TNBC). A recent study explored the use of conjugated polymer nanoparticles (CPNs) as photosensitizers for targeted PDT. By attaching unique 2'-Fluoropyrimidines-RNA-aptamers to the surface of CPNs, the researchers aimed to improve the selectivity of the treatment for TNBC cells. Results showed that CL4, sTN29, and sTN58 aptamers were effective in selective tumor targeting and improving PDT efficacy in TNBC cells.

Sweet light o? mine: Photothermal and photodynamic inactivation of tenacious pathogens using conjugated polymers

Rodrigo A. Ponzio et al.

Summary: The increasing pandemic of antibiotic resistant pathogens has resulted in a rising number of untreated infections. Innovative antibiotics are in short supply globally, which further contributes to the emergence and spread of drug resistant microbes. In this paper, the potential of light-mediated therapies, particularly using conjugated polymers (CPs), for combating multidrug-resistance microbes is discussed. CPs possess unique characteristics that make them a powerful class of photosensitizer/photothermal agent materials. The challenges and needs in evaluating the efficiency of phototherapies are also addressed.

Summary: This study develops a nanomedicine-based cancer sono-immunotherapy that utilizes a semiconducting polymer to generate high levels of 1O2 and conjugates immunomodulators for targeted tumor treatment. The therapy not only reduces tumors and enhances tumor immunogenicity, but also releases immunomodulators specifically at the tumor site. It shows effective antitumor efficacy and reduces immune-related adverse events in animal models.

Antibacterial Sonodynamic Therapy: Current Status and Future Perspectives

Jayishnu Roy et al.

Summary: Multidrug-resistant bacteria have emerged in both community and hospital settings due to antibiotics misuse, leading to efforts to explore alternate antibacterial therapies like sonodynamic therapy (SDT) based on physical mechanisms. SDT involves exposing tissues to sensitizing chemical and low-intensity ultrasound, producing reactive oxygen species for treating bacterial infections, with potential for treating deep-seated infections. Efforts are being made to improve the bioavailability of sonosensitizers using targeted delivery agents like nanoparticle composites, liposomes, and microbubbles.

Summary: Photodynamic inactivation of microorganisms (aPDI) is an effective method to destroy antibiotic-resistant microbial isolates, but research on different photosensitizers, target microorganisms, and light delivery systems mostly shows efficacy in vitro models. Investigating different bacterial subpopulations' responses to aPDI is essential to identifying molecular features driving antimicrobial photodynamic inactivation effectiveness.

Summary: Fluorescent conjugated polymers (CPs) have garnered significant interest in biosensing due to their high fluorescence, tunable bandgap, and good biocompatibility. The efficient binding of CPs to biological targets is crucial for obtaining the desired optical responses, which is largely driven by effective surface interactions. These interactions can lead to changes in the spatial conformation and distribution of CPs, altering their optical properties and enabling diverse bioapplications such as cell imaging, imaging-guided detection, and photodynamic therapy.
