Mechanisms of folate metabolism-related substances affecting Staphylococcus aureus infection

Transcription-coupled DNA repair underlies variation in persister awakening and the emergence of resistance

Summary: Persisters are a type of temporarily antibiotic-tolerant variants in bacterial populations, causing relapsing infections. This study reveals the repair mechanism of oxidative DNA damage upon persister state exit and its relationship with antibiotic resistance development at the molecular level. Results show that persisters accumulate oxidative DNA damage during fluoroquinolone treatment, which is repaired through nucleotide excision repair. Most persisters can repair the damage and have a mutation rate equal to the untreated population, but the promutagenic factor Mfd increases the mutation rate in a subpopulation of persisters. These findings provide insights into the molecular mechanisms of persister survival and link persistence to resistance development.
