Sono-promoted drug penetration and extracellular matrix modulation potentiate sonodynamic therapy of pancreatic ductal adenocarcinoma

Summary: Pancreatic ductal adenocarcinoma (PDAC) exhibits difficulty in drug penetration due to the highly-dense extracellular matrix (ECM). Researchers developed a sono-responsive polymeric perfluorohexane (PFH) nanodroplet to achieve deep drug penetration in PDAC through ultrasonic exposure and ECM modulation for potent sonodynamic therapy (SDT). The PFH nanodroplets were successful in alleviating the physiological barrier of PDAC and showed promising treatment effects.

Biological evaluation of novel gemcitabine analog in patient-derived xenograft models of pancreatic cancer

Summary: The objective of this study was to modify Gem into a more stable form called 4-(N)-stearoyl-gemcitabine (4NSG) and evaluate its therapeutic efficacy in patient-derived xenograft (PDX) models from Black and White patients with pancreatic cancer (PCa). The study found that 4NSG-SLN significantly improved the pharmacokinetic profile of Gem and reduced tumor growth in PDX mice bearing Black and White PCa tumors compared to GemHCl.

Summary: Pharmaceuticals are widely recognized as potentially hazardous to aquatic ecosystems and have been detected in various water sources. Exposure to chemotherapy drugs gemcitabine and paclitaxel in aquatic environments caused developmental toxicity in zebrafish embryos, affecting survival, hatching rate, morphology, and body length. The exposure also disturbed the antioxidant defense system, leading to increased ROS, and affected the expression of inflammation-related, endoplasmic reticulum stress-related, and autophagy-related genes.

Derivation of Human Salivary Epithelial Progenitors from Pluripotent Stem Cells via Activation of RA and Wnt Signaling

Mechanism Study on Nanoparticle Negative Surface Charge Modification by Ascorbyl Palmitate and Its Improvement of Tumor Targeting Ability

Summary: The polarity and density of surface charge influence the immune clearance and cellular uptake of lipid nanoparticles (LNPs). in this study, ascorbyl palmitate (AsP) was used as a negatively charged lipid to modify the surface charge. The amount and distribution of AsP on the LNPs surface were correlated with the zeta potentials. At lower ratios, AsP distributed homogeneously in LNPs and formed strong hydrogen bonds with phosphatidylcholine (PC), promoting LNPs' stability. However, at higher ratios, excessive AsP changed the interaction modes and altered the properties of LNPs.
