MBNL1 drives dynamic transitions between fibroblasts and myofibroblasts in cardiac wound healing

Darrian Bugg et al.

Summary: The study found that MBNL1 plays a key role in controlling the plasticity of cardiac fibroblast state. Overexpression of MBNL1 in cardiac fibroblasts transitions them into myofibroblasts, promoting scar formation, while loss of MBNL1 limits scar production as fibroblasts are unable to transition into myofibroblasts.

Idiopathic Pulmonary Fibrosis: An Update on Pathogenesis

Qianru Mei et al.

Summary: Idiopathic pulmonary fibrosis (IPF) is a progressive and fatal lung disease with limited understanding of its pathogenesis. Recent studies suggest that sustained lung epithelial injury and fibroblast differentiation play key roles in the development of IPF. Current treatments can slow disease progression but cannot cure it.

S100A12 as Biomarker of Disease Severity and Prognosis in Patients With Idiopathic Pulmonary Fibrosis

Yupeng Li et al.

Summary: This study found that S100A12 might be an efficient disease severity and prognostic biomarker in patients with IPF. It is highly expressed in monocytes, but downregulated in lung tissue. Ten genes interacting with S100A12 and the co-expressed genes S100A8 and S100A9 are significantly associated with lung function, survival rates, and survival time in IPF patients. The prognostic predictive value of S100A12 is superior to S100A8 and S100A9 in IPF patients.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

The Effect of Hypoxia on the Expression of CXC Chemokines and CXC Chemokine Receptors-A Review of Literature

Jan Korbecki et al.

Summary: Hypoxia plays a crucial role in the tumor microenvironment, affecting cancer processes. Changes in CXC chemokine expression induced by hypoxia have implications for angiogenesis, lymphangiogenesis, and recruitment of various cells to the tumor niche. Targeting the CXC chemokine system with drugs has potential in cancer therapy.

CD4+T cells in ageing-associated interstitial lung abnormalities show evidence of pro-inflammatory phenotypic and functional profile

Carlos Machahua et al.

Summary: The study found that in ILA patients, the number of Naive CD4+T cells increased, while the number of central and effector memory CD4+T cells decreased. CD4+T cells in ILA patients were highly proliferative and showed excessive functional activity, likely related to the loss of KLRG1 expression.

Blood Transcriptomics Predicts Progression of Pulmonary Fibrosis and Associated Natural Killer Cells

Yong Huang et al.

Summary: Using longitudinal gene expression changes, a predictor for future FVC decline in IPF patients was identified, with high sensitivity and specificity across independent cohorts. Pathway analyses revealed downregulation of pattern recognition receptor pathways and upregulation of mTOR pathways. Cellular deconvolution using single-cell RNA-sequencing data identified natural killer cells as significantly correlated with progression in IPF.

Inhibition of FGF-FGFR and VEGF-VEGFR signalling in cancer treatment

Guihong Liu et al.

Summary: The FGF-FGFR signaling pathway and the VEGF-VEGFR signaling pathway play important roles in the treatment of tumors, and inhibitors targeting these two pathways can optimize therapeutic effects and expand the antitumor range. In addition, biomarkers should be developed in the future to predict efficacy, and combination with immune checkpoint inhibitors is a promising direction.

Genomic analysis uncovers prognostic and immunogenic characteristics of ferroptosis for clear cell renal cell carcinoma

Dan Bai et al.

Summary: This study systematically investigated the characteristic patterns of ferroptosis in ccRCC and its interactions with the tumor microenvironment. High ferroptosis scores were associated with poor prognosis, increased T cell infiltration, elevated immune and stromal scores, and enhanced response to anti-CTLA4 immunotherapy, while low ferroptosis scores were linked to enhanced tumor purity and amino acid and fatty acid metabolism pathways. Evaluation of ferroptosis patterns may help oncologists develop personalized immunotherapeutic strategies.

Investigation of a Hypoxia-Immune-Related Microenvironment Gene Signature and Prediction Model for Idiopathic Pulmonary Fibrosis

Xinyu Li et al.

Summary: This study developed a hypoxia-immune-related prediction model for the prognosis of IPF, identifying high hypoxia and high immune status as risk factors for poor prognosis in IPF patients. Specific immune cells were found to play key roles in the hypoxia-immune-related microenvironment, and a prediction model based on one protective and nine risk genes was established. The model showed significant applicability in peripheral whole blood, peripheral blood mononuclear cell, and lung tissue of IPF patients in independent validation cohorts.
