Redox-Neutral Ruthenium(II)-Catalyzed Enol-Directed Arene C-H Alkylation with Maleimides

Summary: An enol-assisted regioselective arene C-H alkylation with maleimides is developed under redox-neutral ruthenium-(II) catalysis, offering a wide variety of valuable 3-aryl succinimides including amino acid embedded frameworks in good to excellent yields. The products were also aromatized to produce synthetically useful resorcinol-based biaryls. Mechanistic studies support an organometallic pathway with a reversible C-H metalation step for this reaction.

Palladium-Catalyzed Defluorinative Coupling of Difluoroalkenes and Aryl Boronic Acids for Ketone Synthesis

Summary: In this manuscript, a novel palladium-catalyzed coupling reaction between gem-difluoroalkenes and aryl boronic acids is reported, which yields bioactive indane-type ketones with an all-carbon alpha-quaternary center. The mechanism involves a water-induced defluorination and cross-coupling cascade pathway, supported by characterization and stoichiometric reactions of the key intermediates RCF2PdII. The use of vinyl difluoromethylene motif as an in situ carbonyl precursor is unprecedented in transition-metal-catalyzed coupling reactions, and it is expected to spark broad interest in ketone synthesis, fluoroalkene functionalization, and design of new synthetic protocols based on the unique reactivity of difluoroalkyl palladium(II) species.

Cu/TBHP mediated tetrahydroquinoline synthesis in water via oxidative cyclization reaction

Summary: A series of tetrahydroquinoline derivatives were synthesized through Cu(OAc)(2)/TBHP assisted oxidative cyclization reaction. The reactions were carried out in aqueous media under mild conditions, achieving dual functionalization of C(sp(3))-H and C(sp(2))-H bonds in one-pot operation. TBHP assisted synthesis of tetrahydroquinoline analogues was also successfully performed in the absence of copper catalyst.

Photoinduced Electron Donor-Acceptor Complex-Mediated Radical Cascade Involving N-(Acyloxy)phthalimides: Synthesis of Tetrahydroquinolines

Summary: We proposed a novel disconnection approach to synthesize fused tetrahydroquinolines through a visible light-mediated radical (4 + 2) annulation reaction between alkyl N-(acyloxy)-phthalimides and N-substituted maleimides using DIPEA as an additive. The reaction involves a photoactivated electron donor-acceptor complex between alkyl NHPI esters and DIPEA, and regioselective tetrahydroquinolines were obtained. This metal-and catalyst-free methodology exhibits a wide scope, good functional group tolerance, and detailed mechanistic investigations were performed.

Palladium-Catalyzed [3+2] Annulation of Aromatic Amides with Maleimides through Dual C-H Activation

Summary: A palladium-catalyzed [3 + 2] annulation of substituted aromatic amides with maleimides is described, providing tricyclic heterocyclic molecules in good to moderate yields through weak carbonyl chelation. The reaction proceeds via a dual C-H bond activation, first selectively at the benzylic position and then at the meta position, resulting in a five-membered cyclic ring. The use of external ligand Ac-Gly-OH is crucial for the success of this protocol. A plausible reaction mechanism has been proposed for the [3 + 2] annulation reaction.

Acylsilane Directed Rh-Catalyzed Arene C-H Alkylation with Maleimides and Visible-Light-Induced Siloxycarbene-Amide Cyclization: [3+2] Carbo-Annulation in Ru Catalysis

Summary: We demonstrate the Rh-catalyzed arene C-H bond alkylation with acylsilane directing group and maleimides. The resulting products are utilized for the synthesis of new tricyclic gamma-lactams through visible-light-induced intramolecular siloxycarbene-amide cyclization. Additionally, we also show the [3 + 2] carbo-annulation of maleimides and aroylsilanes using Ru-catalysis.

Synthesis of Naphthalimides through Tandem Pd(II)-Catalyzed C(sp3)-H Oxidation and Diels-Alder Reaction Using a Transient Directing Group Strategy

Summary: A new approach for the synthesis of naphthalimides via a tandem reaction of o-methylbenzaldehydes and maleimides is developed in this work. The reaction involves Pd(II)-catalyzed benzylic C(sp(3))-H oxidation using an amino acid as the transient directing group and Diels-Alder reaction. The subsequent dehydration forms naphthalimides, allowing for easy access to a range of naphthalimides with a variety of substituents.

Summary: In the past 15 years, there has been increasing research interest in using light to promote organic transformations. While [2 + 2] cycloadditions are usually performed photochemically, limited literature examples exist on the reaction between olefins and maleimides, particularly focusing on N-alkyl maleimides or using metal catalysts for visible-light driven reactions of N-aryl maleimides. This study identifies the differences in reactivity between N-alkyl and N-aryl maleimides and provides optimized protocols for the reactions using different irradiation wavelengths and photosensitizers.

Overcoming the Challenges toward Selective C(6)-H Functionalization of 2-Pyridone with Maleimide through Mn(I)-Catalyst: Easy Access to All-Carbon Quaternary Center

Summary: An earth-abundant and inexpensive Mn(I)-catalyzed alkylation of 2-pyridone with maleimide has been reported for the first time, which demonstrates an unexpected rearrangement in the presence of acetic acid and provides a unique class of compounds. This methodology shows good tolerance towards various functional groups, delivering the alkylated products in moderate to excellent yields.

Ligand-Enabled [3+2] Annulation of Aromatic Acids with Maleimides by C(sp3)-H and C(sp2)-H Bond Activation

Summary: This paper describes a palladium-catalyzed [3+2] annulation reaction of substituted benzoic acids with maleimides, leading to tricyclic heterocyclic molecules with a free carboxylic group in a highly atom- and step-economical manner. The reaction involves dual C-H bond activation, including C(sp(3))-H activation at the benzylic position and C(sp(2))-H bond activation at the meta position of substituted aromatics. The success of this protocol relies on the presence of an external ligand, MPAA. The reaction exhibited a broad substrate scope and high efficiency, providing a straightforward method for synthesizing succinimide-fused 9,10-dihydrophenanthrenes. Additionally, we also developed a reaction using acenaphthylene as the relay, allowing easy access to dibenzo[j,l]fluoranthenes.

Summary: A catalytic system has been discovered for the intramolecular C-H amidation of N-phenoxy acetamide derivatives. The use of a cobalt catalyst and maleimide as a transient mediator enables the olefin-mediated ortho C-H functionalization. Mechanistic and DFT studies provide support for the proposed mechanism. The products derived from this methodology have been demonstrated as ligands in the C-H olefination reaction.

Copper(I)-Mediated Cascade Annulation via Dual C-H/C-H Activation: Access to Benzo[a]carbazolic AEEgens

Summary: A Cu(I)-mediated cascade cyclization/annulation of unprotected o-alkynylanilines with maleimides in one pot is developed, providing fused benzo[a]carbazoles with free NH skeletons. The annulated products exhibit fluorescence emission in the range of 485-502 nm, with a large Stokes shift and fluorescence lifetime of around 17 ns. Cell viability assays confirm the biocompatibility of the annulated products and their potential for cell imaging applications.
