Role of epigenetics in OSCC: an understanding above genetics

Summary: Oral cavity cancer, also known as oral squamous cell carcinoma, is a type of head and neck cancer that primarily develops from squamous cells. While molecular markers for oral cavity cancer are known, the exploration of epigenetic signatures is limited. Recent studies have revealed the interdependency of epigenetic and genetic alterations in various cancers, making epigenetic changes an attractive target for cancer therapeutics. DNA methylation and histone modifications are the primary epigenetic alterations in cancer, which can be used for early patient detection and prognostication. This review provides an overview of the epigenetic perspective on the etiology, biomarkers, and potential drugs for the predictive diagnosis and treatment of oral squamous cell carcinoma.

Hallmarks of Cancer: New Dimensions

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Nrf2 was found to enhance glycolysis and glutathione synthesis, thus contributing to radioresistance through metabolic reprogramming. Furthermore, phosphorylated Nrf2 expression was correlated with pathological response to chemoradiotherapy and prognosis in advanced OSCC patients.

Targeting CES2 may provide a potential therapeutic strategy for OSCC.

Summary: The poor drug efficacy is a primary cause of treatment failure in oral squamous cell carcinoma (OSCC). Despite efforts in developing therapeutic strategies for OSCC, effective drugs are still lacking and the 5-year survival rate of patients with OSCC is low. The study investigated the apoptotic effect of lycorine hydrochloride (LH) on OSCC HSC-3 cell line, demonstrating that LH induced cell apoptosis and cell cycle arrest, inhibited cell proliferation, and activated ROS-mediated mitochondrial apoptotic pathway and JNK signaling pathway in OSCC cells, suggesting LH as a potential drug candidate for anti-OSCC therapy.

However, low expression of POLB is associated with advanced tumor stage and poor overall survival in OSCC patients, indicating its potential role in regulating cell growth through cell cycle modulation and chromosomal instability. Studies also suggest that POLB is important in maintaining karyotypic stability in OSCC cells, affecting outcomes and survival rates.

The selective toxicity of superparamagnetic iron oxide nanoparticles (SPIONs) on oral squamous cell carcinoma (OSCC) by targeting their mitochondria
