Characteristics of human papillomavirus infection among oropharyngeal cancer patients: A systematic review and meta-analysis

Consensuses, controversies, and future directions in treatment deintensification for human papillomavirus-associated oropharyngeal cancer

Summary: Oropharyngeal cancer (OPC) is the most common cancer caused by human papillomavirus (HPV) infection in the United States. Identifying the optimal deintensification strategy for HPV-positive OPC has been a key focus for academic head-and-neck surgery, radiation oncology, and medical oncology. The first generation of randomized chemotherapy deintensification trials did not change the standard of care, leading to concerns about de-escalation feasibility. National database studies show that a significant number of patients receive nonstandard de-escalated treatments. Synthesizing multidisciplinary deintensification data and treatment standards is important to reinforce best practices and improve patient outcomes.

Analysis of a hit-and-run tumor model by HPV in oropharyngeal cancers

Summary: Some cancers, including certain anogenital and head and neck cancers, may be caused by viruses such as human papilloma virus (HPV). The presence of the virus in tumors has usually been used to establish causality, but this study suggests that the absence of viral genes from tumors does not exclude the possibility of viral causation. By deleting the viral oncogene E7 using CRISPR, the researchers found that tumors were eliminated, but the deleted tumors recurred over time and developed new mutations, some of which were already present in tumors without HPV.

Human Papillomavirus and Rising Oropharyngeal Cancer Incidence in the United States

Summary: Increases in the incidence and survival of oropharyngeal cancers in the United States since 1984 are attributed to HPV infection. This study examined 271 oropharyngeal cancer patients and found that HPV-positive patients had a significantly longer survival period. The incidence of HPV-positive oropharyngeal cancers is expected to exceed that of cervical cancers by 2020.

Summary: This study found that the prevalence of oropharyngeal squamous cell carcinomas (OPSCCs) driven by HPV infection is increasing in Northeastern Italy. The study also suggests that the prevalence of HPV infection impacts the positive predictive value of p16(INK4a) overexpression. Therefore, each institution should consider the subsite-specific prevalence rates of HPV-driven OPSCC when using p16(INK4a) overexpression as a surrogate marker for transforming HPV infection.
