Deciphering postnatal limb development at single-cell resolution

Manman Gao et al.

Summary: A systematic study was conducted using single-cell RNA sequencing technique to characterize 19,952 individual cells from murine hindlimbs at 4 postnatal stages. The study revealed the presence of candidate progenitor sub-clusters in articular cartilage and enthesis, as well as three cellular developmental branches in the growth plate. The transcriptomes and developmental patterns were extensively explored, and the implications for osteoarthritis were discussed. Overall, these findings broaden our understanding of postnatal limb developmental biology and the interconnections between limb development, remodeling, and regeneration.

Osteoarthritis Pain

Huan Yu et al.

Summary: Joint pain is a major symptom of osteoarthritis and can significantly affect joint function and quality of life. Current treatment strategies for osteoarthritis pain are often unsatisfactory and can have harmful side effects. Therefore, understanding the mechanisms behind joint pain is crucial for the development of more effective and specific drugs.

Characteristics of sensory innervation in synovium of rats within different knee osteoarthritis models and the correlation between synovial fibrosis and hyperalgesia

Li Zhang et al.

Summary: This study investigated the characteristics of sensory innervation in different KOA rat models and found a positive correlation between the degree of synovial fibrosis and the pain sensitivity of the KOA model rats. Surgical modeling, especially the ACLT method, is recommended for KOA pain research.

Infrapatellar Fat Pad-Synovial Membrane Anatomo-Fuctional Unit: Microscopic Basis for Piezo1/2 Mechanosensors Involvement in Osteoarthritis Pain

Aron Emmi et al.

Summary: The study investigates the differences in cell marker expression and distribution between healthy and osteoarthritis (OA) patients, shedding light on the mechanisms and potential targets for OA pain. The findings highlight the importance of the Infrapatellar Fat Pad-Synovial Membrane anatomical unit and its role in triggering and perpetuating OA disease.

Sensory Neuron-Specific Deletion of Tropomyosin Receptor Kinase A (TrkA) in Mice Abolishes Osteoarthritis (OA) Pain via NGF/TrkA Intervention of Peripheral Sensitization

Insug O-Sullivan et al.

Summary: The NGF/TrkA signaling pathway plays a crucial role in osteoarthritis pain, involving cellular plasticity in peripheral sensory neurons and angiogenesis in synovial cells. In mice with sensory-neuron-specific deletion of TrkA, the OA-related joint pain was significantly improved.

VEGF-A/VEGFR-1 signalling and chemotherapy-induced neuropathic pain: therapeutic potential of a novel anti-VEGFR-1 monoclonal antibody

Laura Micheli et al.

Summary: Studies in murine models suggest that VEGF-A and its receptor VEGFR-1 play a crucial role in chemotherapy-induced neuropathic pain, mediating pain transmission through communication between astrocytes and neurons. Blocking VEGFR-1 may help alleviate neuropathic pain by disrupting this interaction.

Yvonne D'Arcy et al.

Summary: Osteoarthritis is a common difficult-to-treat condition, with the goal of alleviating symptoms such as pain and loss of function through pharmacologic treatments. In addition to traditional medication options, antibodies directed against nerve growth factor are a new class of agents under clinical investigation for the treatment of osteoarthritis.

Expression of VEGF-A Signaling Pathway in Cartilage of ACLT-induced Osteoarthritis Mouse Model

Jia-jia Qian et al.

Summary: ACLT surgery can lead to time-dependent degeneration of cartilage in mice. Increased ADAMTS5-positive cells and decreased COL-II in OA cartilage, along with angiogenesis and inflammation, were important pathological changes. The expression of the VEGF-A/VEGFR2 signaling pathway was correlated with the degree of KOA.
