Empagliflozin protects mice against diet-induced obesity, insulin resistance and hepatic steatosis

Bernhard Radlinger et al.

Summary: This study aimed to test whether empagliflozin can prevent weight gain and metabolic dysfunction in a mouse model of diet-induced obesity and insulin resistance. The results showed that empagliflozin supplementation prevented diet-induced metabolic abnormalities and protected mice from obesity, insulin resistance, and fatty liver. Furthermore, empagliflozin supplementation improved mitochondrial morphology in skeletal muscle.

Summary: This study aimed to investigate the impact of Empagliflozin on SKM function and mitochondrial function in an established HFpEF rat model. The results showed that Empagliflozin treatment improved SKM function and contractility, reduced intramuscular lipid content, and had positive effects on mitochondrial function.

Excessive exercise training causes mitochondrial functional impairment and decreases glucose tolerance in healthy volunteers

Summary: Exercise training positively affects metabolic health, but excess exercise may lead to reduced mitochondrial function and impaired glucose tolerance. Studies have shown that world-class endurance athletes have issues with glucose control compared to matched control groups.

Lipids activate skeletal muscle mitochondrial fission and quality control networks to induce insulin resistance in humans

Christopher L. Axelrod et al.

Summary: The study found that lipid infusion in human skeletal muscle activates mitochondrial fission and quality control, reduces mitochondrial membrane potential, increases lipid droplet formation, dilated cristae, and presence of autophagic vesicles. Additionally, lipid infusion suppresses peripheral glucose disposal and hepatic insulin sensitivity.

Summary: The study demonstrates that DAPA treatment can prevent mitochondrial swelling and normalize mitochondrial size in diabetic animals, reverse the decline in mtDNA copy number, increase gene expression in liver tissue, restore mitochondrial respiratory control ratio, and reduce lipid peroxidation products levels in liver mitochondria.

Reduced metabolic capacity in fast and slow skeletal muscle via oxidative stress and the energy-sensing of AMPK/SIRT1 in malnutrition

Takumi Hirabayashi et al.

Summary: The study found that malnutrition leads to a decrease in metabolic capacity in both fast and slow muscles, affecting muscle mass and body weight, with increased oxidative stress and inhibition of SIRT1. These results suggest that malnutrition impairs metabolic capacity in both types of muscles through SIRT1 inhibition induced by increased oxidative stress.
