Leucine-Rich Alpha-2 Glycoprotein 1 Accumulates in Complicated Atherosclerosis and Promotes Calcification

CD163+macrophages restrain vascular calcification, promoting the development of high-risk plaque

Summary: Vascular calcification (VC) and atherosclerosis coexist, but it is unclear why rupture-prone high-risk plaques do not typically calcify extensively. In this study, the researchers found that CD163+ macrophages, which are involved in atherosclerosis, have an inverse correlation with VC in human arteries. They also discovered that these macrophages inhibit VC through NF-KB-induced hyaluronan synthase (HAS), an enzyme that plays a role in the formation of the extracellular matrix. These findings provide insights into the mechanism by which CD163+ macrophages promote high-risk plaque development by inhibiting VC.

Modulation of transforming growth factor-β-binduced kidney fibrosis by leucine-rich α-2 glycoprotein-1

Summary: The abnormal expression of secreted leucine-rich alpha-2 glycoprotein-1 (LRG1) is associated with the progression of chronic kidney diseases, promoting kidney fibrosis through enhanced endothelial transforming growth factor-beta (TGF-beta) signaling. LRG1 expression increases in glomerular endothelial cells and the tubulointerstitial compartment. In mouse models, loss of LRG1 attenuated kidney fibrosis, while overexpression exacerbated fibrosis. These findings suggest that therapeutic targeting of LRG1 may effectively halt the progression of kidney fibrosis in chronic kidney disease.

Programmed cell death in atherosclerosis and vascular calcification

Summary: The concept of cell death has expanded to include various forms such as necroptosis, pyroptosis, autophagy, and ferroptosis, in addition to apoptosis and necrosis. These different modes of cell death play critical roles in all aspects of life and have diverse implications in diseases. Atherosclerosis (AS) and vascular calcification (VC) are major causes of high morbidity and mortality in cardiovascular disease. Despite advancements in understanding the signaling pathways associated with AS and VC, the exact molecular basis remains unknown. This article reviews the molecular mechanisms underlying cell death and its implications for AS and VC, which may lead to the development of promising therapeutic strategies.

Summary: In this study, the authors investigated the role of leucine-rich alpha-2-glycoprotein 1 (LRG1) in the apoptosis and autophagy of H9c2 cells in the context of acute myocardial infarction (AMI). They found that LRG1 promotes apoptosis and autophagy under hypoxic conditions, and the effects can be reversed by downregulating LRG1 or knocking down hypoxia-inducible factor-1 alpha (HIF-1 alpha).
