pH- and Temperature-responsive Hydrogels Based on Tertiary Amine-modified Polypeptides for Stimuli-responsive Drug Delivery

Summary: pH- and temperature-responsive hydrogels based on mPEG-polypeptide block copolymers were developed in this study. The hydrogels exhibited an alpha-helix-to-beta-sheet transition at increased pH and underwent thermo-induced sol-gel phase transitions at elevated polymer concentrations, dependent on the pH. They demonstrated good histocompatibility and almost complete degradation in the subcutaneous layer of mice. The doxorubicin-loaded hydrogels showed pH-responsive drug release profiles with rapid release at acidic pH and sustained release at neutral pH, suggesting their potential as environment-responsive depots for drug delivery.

Effect of Hofmeister Salts on the LCST of Poly(diethyl vinylphosphonate) and Poly(2-vinylpyridine-block-diethyl vinylphosphonate)

Summary: This study investigates the effect of Hofmeister anions on the cloud point of poly(diethyl vinylphosphonate) (PDEVP) homopolymers and poly(2-vinylpyridine-block-diethyl vinylphosphonate) (P(2VP-b-DEVP)) block copolymers. The results show that the P2VP block does not affect the cloud point temperature (T-CP), but a linear increase in T-CP is observed when the block copolymer is used in the presence of copper(II) or iron(III). These findings have potential applications in sensors or wastewater purification.

Chirality-Governed UCST Behavior in Polypeptides

Design and synthesis of amphiphilic alternating peptides with lower critical solution temperature behaviors

Summary: Amphiphilic peptides with repeating hydrophilic and hydrophobic units were synthesized using Ugi's four-component condensation reaction and their turbidity in aqueous solutions were measured at different temperatures. The study found that the alternating peptides showed thermoresponsive behavior, with peptides containing repeating hydrophilic and hydrophobic units displaying upper critical solution temperature (UCST) behavior, and those with large enough hydrophobic units exhibiting lower critical solution temperature (LCST) behavior. Additionally, thermoresponsive hydrogels were prepared using the alternating peptides as cross-linking points, and their swelling in water was temperature-dependent.

Water-assisted and protein-initiated fast and controlled ring-opening polymerization of proline N-carboxyanhydride

Emerging opportunities in bioconjugates of Elastin-like polypeptides with synthetic or natural polymers

This review focuses on the natural structural proteins, particularly those characterized by repeat peptide sequences, which have been extensively studied for the design of biomimetic materials. Elastin and its main repeat sequences have been the subject of intense research for over 50 years, leading to the field of elastin-like polypeptides (ELPs). ELPs are currently widely investigated in various applications, including protein purification, tissue engineering, and drug delivery, with some technologies based on ELPs being explored by start-up companies. The review summarizes pioneering contributions on ELPs, progress in their genetic engineering, and understanding of their thermal behavior and self-assembly properties. Additionally, the review highlights the opportunities that remain unexplored in the conjugation of ELPs with other synthetic macromolecules as covalent hybrids, statistical, graft, or block copolymers.

The synthesized diblock copolymer exhibited gel-to-sol UCST transition behavior in an aqueous solution, with the transition temperature window easily adjusted by changing the copolymer concentration or length of the PBnLT block. The disassembly of initial beta-sheet nanoassemblies caused nanofibril transformation to spherical aggregates with increasing temperature, leading to a gel-to-sol UCST transition. This result inspires a brand-new strategy for the structural design and functional control of materials.

Additionally, membrane permeability was found to increase with the log P value of the fluorophores, suggesting electrical charge and log P value are important factors for permeability.

Different Folding States from the Same Protein Sequence Determine Reversible vs Irreversible Amyloid Fate

Summary: The propensity of proteins to self-assemble into amyloid fibrils with a shared cross-beta architecture is common, but the irreversibility and stability of amyloid fibrils make amyloid-related diseases difficult to treat. Recent studies suggest that certain proteins with low-complexity domains may form labile amyloids. Research shows that reversible and irreversible amyloid fibrils formed from the same full-length protein sequence have differences in core structures and beta-sheet arrangements, highlighting the role of protein folding states in regulating amyloid lability and reversibility.

Summary: Hybrids of synthetic polymers and biopolymers, such as cationic ELPs, were successfully obtained by chemoselectively modifying methionine residues to introduce alkyne groups and coupling cationic oligomers through Huisgen azide-alkyne cycloaddition reaction. The characterization of these hybrid cationic ELPs by H-1 NMR, zeta-potential and SEC analyses showed their purity, degree of functionalization, overall charge, and molar mass. Preliminary biological tests demonstrated the efficient internalization of genetic material into living cells using these hybrid cationic ELPs.
