Alzheimer's disease: a mini-review for the clinician

Interaction of Exogenous Butyrylcholinesterase with β-Amyloid Plaques in 5XFAD/Butyrylcholinesterase-Knockout Mouse Brain

Summary: In Alzheimer's disease (AD) and amyloid models such as the 5XFAD mouse, butyrylcholinesterase (BChE) is associated with beta-amyloid (Aβ) plaques and has unique biochemical properties. This study found that BChE bound to Aβ plaques in a mouse model exhibited similar properties to those seen in AD, suggesting a conformational change when bound to Aβ plaques but not neurons. The findings indicate the potential role of BChE in the pathogenesis of AD and provide insights into the binding mechanism of BChE to Aβ plaques.

Catalytic Amyloids as Novel Synthetic Hydrolases

Summary: Amyloids are supramolecular assemblies composed of polypeptides, which have been traditionally associated with pathological conditions but are now looked at as alternatives for designing novel active bionanomaterials. Recent findings have shown that amyloid fold can serve as catalytic scaffolds, leading to the development of catalytic amyloids with hydrolytic activities. These artificial hydrolases, emerging upon self-assembly of small peptides into amyloids, could provide a stable and attractive alternative for future design of synthetic hydrolases with diverse applications in industries such as in situ decontamination of xenobiotics.

Monitoring the early aggregatory behaviour and size of Aβ1-42 in the absence & presence of metal ions using dynamic light scattering
