In Search of Biomarkers to Guide Interventions in Autism Spectrum Disorder: A Systematic Review

Mara Parellada et al.

Summary: The aim of this study was to evaluate response biomarkers correlated with autism spectrum disorder (ASD) symptoms. A systematic review was conducted and 280 articles were included, reporting on 940 biomarkers. However, the studies showed high heterogeneity and there is currently no sufficient evidence for response biomarkers in ASD clinical trials.

Altered kynurenine pathway metabolite levels in toddlers and preschool children with autism spectrum disorder

Ayhan Bilgic et al.

Summary: This study aimed to investigate kynurenine pathway metabolite levels, enzyme activities, and neuroprotective index in toddlers and preschool children with ASD. The results showed significantly higher serum 3-hydroxykynurenine and kynurenic acid concentrations in the ASD group compared to controls, indicating a potential role of the kynurenine pathway in the etiopathogenesis of ASD in early childhood.

The immune-kynurenine pathway in social anxiety disorder

Mary Butler et al.

Summary: The study found that patients with Social Anxiety Disorder (SAD) had elevated levels of kynurenic acid (KYNA) and KYNA/KYN ratio compared to healthy controls. SAD patients with a history of past suicide attempts showed higher kynurenine (KYN) levels and KYN/TRYP ratio. Additionally, SAD males had significantly lower levels of anti-inflammatory cytokine IL-10 compared to healthy males.

Genetics of glutamate and its receptors in autism spectrum disorder

Sabah Nisar et al.

Summary: Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by deficits in social interaction skills, impaired communication, and repetitive and restricted behaviors. Research has shown that altered glutamate metabolism may be involved in the development of ASD. Imbalance in excitatory and inhibitory neurotransmitters in the glutamatergic system has been observed in pharmacological, behavioral, and imaging studies, suggesting its role in ASD pathology.

Summary: This study investigates the relationship between inflammation and changes in brain structures, and explores the transcriptome-driven functional basis with relevance to mental illness. It finds that genetically determined IL-6 is associated with brain structure and potentially affects areas implicated in developmental neuropsychiatric disorders such as schizophrenia and autism.

Summary: This study found that an oral tryptophan challenge can increase the concentrations of kynurenine, KYNA, and 5-HIAA, which may be elevated in patients with schizophrenia. However, there were no significant differences in baseline levels and stimulation effects between patients with schizophrenia and healthy controls.

Kynurenic acid as a biochemical factor underlying the association between Western-style diet and depression: A cross-sectional study

Heather M. Francis et al.

Summary: Consumption of a Western-style diet high in saturated fat and added sugar is associated with increased depression risk, potentially through altering tryptophan metabolism along the kynurenine pathway and linking inflammation with depression. This study found that urinary levels of the neuroprotective kynurenic acid (KA) were lower with higher intake of the Western-style diet, and KA was significantly associated with depression symptoms independent of inflammation. These findings suggest a role of KA in regulating depression severity in the absence of inflammation.

Influence of glutamate and GABA transport on brain excitatory/inhibitory balance

Sheila M. S. Sears et al.

Summary: An optimally functional brain requires a regulated and balanced combination of excitatory and inhibitory inputs. Imbalance in this interplay, as seen in various neurological disorders and developmental disorders, can lead to dysfunctional signaling and impaired cognitive or motor function. The neurotransmitters glutamate and GABA play a key role in controlling this balance at the cellular level.

Effects of neuroactive metabolites of the tryptophan pathway on working memory and cortical thickness in schizophrenia

Junchao Huang et al.

Summary: This study found that patients with schizophrenia had significantly lower serum levels of KYNA and QUIN, and higher 5-HI/KYNA and QUIN/KYNA ratios compared to healthy controls. Patients with high 5-HI and low KYNA showed better working memory, and higher 5-HI levels were associated with thicker cortex in a specific brain area among patients. The different effects of 5-HI and KYNA on cognitive function suggest potential for novel therapeutic development targeting cognitive impairment in schizophrenia.

The Effects of Maternal Interleukin-17A on Social Behavior, Cognitive Function, and Depression-Like Behavior in Mice with Altered Kynurenine Metabolites

Yuki Murakami et al.

Summary: Viral infection and chronic maternal inflammation during pregnancy have been associated with a higher risk of autism spectrum disorder (ASD). IL-17A and kynurenine, two molecules affected by maternal inflammation, may play a crucial role in the development of ASD-like behavioral abnormalities in offspring by altering metabolism and neuroactive compounds in the fetal brain.
