Summary: The distinct tumor microenvironment (TME) of prostate cancer (PCa) consists of various stromal cells, immune cells, and a dense extracellular matrix (ECM), which collectively promote tumor proliferation and progression. Understanding the components of the prostate TME, including tertiary lymphoid structures (TLSs) and metastasis niches, provides insights into tumor metastasis. Combining the knowledge of TME and emerging therapeutic technologies, several TME-targeted therapies have been developed and tested in clinical trials. This review discusses the components of PCa TME, summarizes TME-targeted therapies, and provides insights into PCa carcinogenesis, progression, and therapeutic strategies.

Knockout of stromal Mcam attenuates orthotopically injected colorectal tumoroid growth and improves survival through decreased tumor-associated macrophage recruitment. The fibroblast MCAM interacts with interleukin-1 receptor 1 to promote macrophage chemotaxis.
