Design of Conjugates Based on Sesquiterpene Lactones with Polyalkoxybenzenes by Click Chemistry to Create Potential Anticancer Agents

Summary: By using click chemistry, a method for synthesizing unique conjugates based on sesquiterpene lactones, namely dehydrocostuslactone and alantolactone, has been developed. The length of the 1,2,3-triazole spacer was varied to expand the structural range of the conjugates. It was found that conjugates based on alantolactone with a long spacer and those containing dehydrocostuslactone with a short spacer exhibited the greatest cytotoxic effect, possibly by causing dissipation of the transmembrane potential of mitochondria and inhibiting glycolysis.

Desloratadine, an FDA-approved cationic amphiphilic drug, inhibits SARS-CoV-2 infection in cell culture and primary human nasal epithelial cells by blocking viral entry

Summary: This study assessed the effectiveness of eight FDA-approved cationic amphiphilic drugs (CADs) against SARS-CoV-2 and found that desloratadine, an antiallergic drug, showed potent antiviral activity not only against SARS-CoV-2 but also against other coronaviruses. It was found that desloratadine targeted an early step of the virus lifecycle and blocked its entry into cells. Experiments on human nasal epithelial cells also demonstrated significant delay in viral RNA production with desloratadine treatment.

Discovery of N-Containing (-)-Borneol Esters as Respiratory Syncytial Virus Fusion Inhibitors

Summary: In this study, a series of potent respiratory syncytial virus (RSV) entry inhibitors with the (-)-borneol scaffold were discovered. The most active compounds, 3b and 5a, showed higher potency than the known antiviral agent Ribavirin. Time-of-addition and temperature shift studies suggested that these compounds inhibit RSV entry by interacting with the viral F protein. Molecular modeling and dynamics confirmed the binding mode of compounds 3b and 5a. These findings highlight the potential of (-)-borneol esters as lead compounds for developing new anti-RSV agents.

Borneol Ester Derivatives as Entry Inhibitors of a Wide Spectrum of SARS-CoV-2 Viruses

Summary: In this study, we investigated the antiviral activity of a home library of monoterpenoid derivatives using pseudoviral systems. We identified borneol derivatives with a tertiary nitrogen atom as potential inhibitors of early viral replication. Further biological tests and studies revealed two lead compounds that showed activity against multiple strains of SARS-CoV-2. Molecular dynamics and docking methods were employed to predict the potential binding sites of the lead compounds, suggesting conserved regions of the glycoprotein S of the virus.

Evidence for distinct mechanisms of small molecule inhibitors of filoviral entry

Despite many potential entry inhibitors, the lack of understanding of their mechanisms of action has contributed to a high failure rate. This report provides molecular insights into multiple mechanisms of action by small molecule inhibitors, offering important guidance for future antiviral drug development.

MolGpKa is a convenient tool for rapid estimation of pK(a) during ligand design.

Identification of SARS-CoV-2 entry inhibitors among already approved drugs

Summary: A high throughput screening assay was developed to identify entry inhibitors for SARS-CoV-2 virus, leading to the discovery of 15 active drugs with specific antiviral properties. Seven of these drugs showed promising levels of activity in inhibiting SARS2 replication in Vero E6 cells, suggesting their potential for further in vivo studies and clinical investigations for COVID-19 treatment.
