Summary: This study aims to evaluate the synergistic effects of shikonin and cisplatin on cisplatin-resistant ovarian cancer cells and investigate the underlying molecular mechanisms. The results showed that the combination of shikonin and cisplatin can effectively reduce the viability of cisplatin-resistant ovarian cancer cells and induce ferroptosis through upregulation of HMOX1 and promotion of Fe2+ accumulation.

NRF2 controls iron homeostasis and ferroptosis through HERC2 and VAMP8

Summary: Enhancing intracellular labile iron pool (LIP) is an effective strategy for inducing ferroptotic death in cancer cells. This study demonstrates that NRF2 regulates iron homeostasis through controlling the expression of HERC2 and VAMP8. Deletion of NFE2L2/NRF2 leads to accumulated apoferritin in autophagosomes, increased LIP, and enhanced sensitivity to ferroptosis. Inhibition of NRF2 in preclinical models has shown the potential of increasing LIP and killing cancer cells through ferroptosis.
