One Stone, Three Birds: A Smart Single Fluorescent Probe for Simultaneous and Discriminative Imaging of Lysosomes, Lipid Droplets, and Mitochondria

Summary: Complex intracellular processes require the cooperation of multiple organelles. Realtime tracking of their interplays is important for understanding biological processes. We report a single fluorescent probe (SFP) that can simultaneously differentiate and visualize three important organelles: mitochondria, lysosomes, and lipid droplets. This smart probe allows for the real-time and simultaneous tracking of the interplays of these organelles for the first time.

A multifunctional fluorescent probe for dual-color visualization of intracellular mitochondria and lipid droplets and monitoring of SO2 in vivo

Summary: This study proposes a SO2-dependent strategy to construct a new functional fluorescence conversion probe MLD-SO2 for two-color visualization of mitochondria and lipid droplets (LDs). The probe selectively targets mitochondria and emits red fluorescence under normal physiological conditions. In the presence of SO2, the probe is converted to a lipophilic structure, targets LDs, and displays strong blue emission. The probe has been successfully used to monitor SO2 changes in zebrafish and mung bean sprouts, making it a powerful tool for studying the interactions between mitochondria and LDs and monitoring SO2 in different organisms.

Summary: A fluorescent biosensor named C-Py was developed with good specificity, low background signal, excellent photostability, low cytotoxicity, and high cellular permeability for tracking LD contact sites with multiple organelles using SIM. The subcellular distribution and dynamic processes of LDs in living cells were observed under SIM with C-Py, revealing contact sites at a resolution below 200 nm. The number of LD-mitochondria contact sites was decreased by lipopolysaccharide induction, suggesting a potential regulatory role of LDs in inflammatory environments.
