Long non-coding RNAs: definitions, functions, challenges and recommendations

John S. S. Mattick et al.

Summary: Genes encoding lncRNAs are abundant in complex organisms and are transcribed by RNA polymerase I, II, and III, as well as processed introns. The classification and annotation of lncRNAs are challenging due to their various functions, isoforms, and interactions with other genes. These lncRNAs evolve rapidly, are cell type-specific, and regulate multiple cellular processes, including gene expression and translation control.

Advances in endogenous RNA pull-down: A straightforward dextran sulfate-based method enhancing RNA recovery

Fabio Desideri et al.

Summary: Detecting RNA/RNA interactions in a given cellular system is essential for understanding the molecular mechanisms of specific RNA molecules. While exogenous methods have been useful, they may fail to reveal biologically meaningful contacts occurring in vivo. Endogenous methods are preferred for the initial identification of partners specifically interacting with selected RNAs.

Identification of Protein-RNA Interactions in Mouse Testis Tissue Using fRIP

Alexis Bailey et al.

Summary: During development, rapid transitions between cell states are crucial for precise and timely differentiation. This can be achieved through post-transcriptional control of gene expression mediated by RNA-binding proteins. Accurately identifying the RNA targets of these proteins at specific stages is essential for understanding their functional role. The study presents an adapted fRIP protocol to identify the in vivo RNA targets of the cytoplasmic RNA-binding protein, YTHDC2, during spermatogenesis. This protocol enables efficient identification of endogenous RNAs and facilitates stage-specific monitoring of developmental changes.

Gene regulation by long non-coding RNAs and its biological functions

Luisa Statello et al.

Summary: Recent studies have shed new light on the biogenesis and functions of long non-coding RNAs (lncRNAs), showcasing their diverse roles in gene regulation and signaling pathways, particularly in the contexts of neuronal disorders, immune responses, and cancer. The discovery of unique biogenesis pathways and subcellular localizations of lncRNAs has opened up potential therapeutic avenues for targeting lncRNAs in various biological and pathophysiological conditions.

RNA-Binding Proteins Hold Key Roles in Function, Dysfunction, and Disease

Sophia Kelaini et al.

Summary: RNA-binding proteins (RBPs) play a crucial role in regulating RNA, and their dysregulation can lead to various diseases including diabetes, cardiovascular disease, cancer, and neurodegenerative diseases. Proper regulation of RNA and RBPs is essential for maintaining good health, as the loss of function can cause pathogenesis.
