Effect of NANOG overexpression on porcine embryonic development and pluripotent embryonic stem cell formation in vitro

Gerelchimeg Bou et al.

Summary: While Nanog is important for maintaining pluripotency in pigs and mice, overexpression of NANOG in early porcine embryos does not improve the efficiency of pluripotent embryonic stem cell clone formation. Therefore, NANOG overexpression is unnecessary for the initial establishment of porcine pluripotent embryonic stem cell clones in vitro.

Changes in OCT4 expression play a crucial role in the lineage specification and proliferation of preimplantation porcine blastocysts

Mingyun Lee et al.

Summary: This study investigated the role of OCT4 in cell lineage specification and embryo proliferation in porcine parthenotes. The results demonstrated that OCT4 plays a crucial role in pluripotency, ICM formation, secondary cell fate decision, and cell proliferation.

NANOG initiates epiblast fate through the coordination of pluripotency genes expression

Nicolas Allegre et al.

Summary: The initiation and regulation of epiblast lineage specification during embryonic development remains unclear. This study shows that coordinated expression of pluripotency markers is crucial for epiblast identity, and that this process is triggered by NANOG activity in a stochastic manner. The findings suggest that these features are likely conserved in human embryos as well.

Species-Specific Enhancer Activity of OCT4 in Porcine Pluripotency: The Porcine OCT4 Reporter System Could Monitor Pluripotency in Porcine Embryo Development and Embryonic Stem Cells

Seung-Hun Kim et al.

Summary: This study investigated the activity and function of the pig OCT4 enhancer in early embryonic development and embryonic stem cells. The findings showed that the pig OCT4 distal enhancer and proximal enhancer were simultaneously activated with different expression patterns during embryo development and the establishment of embryonic stem cells. This research demonstrates the applicability of the porcine OCT4 upstream region-derived dual-fluorescence reporter system for the study of species-specific pluripotency in pig cells.

Comparative analysis of mouse and human preimplantation development following POU5F1 CRISPR/Cas9 targeting reveals interspecies differences

P. Stamatiadis et al.

Summary: POU5F1 is crucial for normal development of mouse and human embryos to the blastocyst stage. Knockout of POU5FI in mice results in compromised embryonic development with loss of inner cell mass and downregulation of gene expression. Similarly, targeted editing of POU5F1 in human embryos leads to developmental impairment, irregular trophectoderm cell layer, and absence of inner cell mass.

SOX2 plays a crucial role in cell proliferation and lineage segregation during porcine pre-implantation embryo development

Mingyun Lee et al.

Summary: The study revealed that SOX2 plays a crucial role in inner cell mass formation and cell proliferation in early-stage porcine embryogenesis. Disruption of SOX2 inhibits the expression of inner cell mass-related genes and reduces total cell number; while overexpression of SOX2 greatly increases blastocyst cell number but decreases the inner cell mass/trophectoderm ratio.

Hypoblast Formation in Bovine Embryos Does Not Depend on NANOG

Claudia Springer et al.

Summary: The role of NANOG in bovine embryo development was studied by knocking out the gene in fibroblast cells and producing NANOG knockout embryos via somatic cell nuclear transfer. The results showed that the FGF/MEK signaling pathway mediated by NANOG is not necessary for hypoblast formation in bovine embryos, suggesting the existence of another unknown pathway regulating hypoblast differentiation.
