Novel therapeutic mechanism of action of metformin and its nanoformulation in Alzheimer's disease and role of AKT/ERK/GSK pathway

Summary: This study investigated the therapeutic mechanism of metformin and its solid-lipid nanoformulation in a rat model of Alzheimer's disease. The results showed that both metformin and its nanoformulation were effective in treating Alzheimer's disease through the AKT-ERK-GSK3 beta-Hyperphosphorylated tau pathway.

Summary: Neurodegenerative diseases and neurodevelopmental disorders are challenging to treat due to the blood-brain barrier. The nose-to-brain pathway offers a promising solution, especially for intranasally administered nanoparticle drugs. Polymeric nanocarriers have shown great potential in improving drug bioavailability in the brain. This review provides an overview of the challenges faced and the contribution of intranasal nano-drug delivery systems in treating CNS conditions.

Brain targeted peptide-functionalized chitosan nanoparticles for resveratrol delivery: Impact on insulin resistance and gut microbiota in obesity-related Alzheimer?s disease

Summary: In this study, Resveratrol (Res) with low solubility was encapsulated in chitosan (CS) cross-linked with sodium tripolyphosphate (TPP) nanoparticles. A brain-targeted peptide (TG) was modified on the nanoparticle surface for specific delivery of Res to the brain and improving cognitive impairments in obesity-related Alzheimer's disease (AD) mice. TG-Res-CS/TPP-NPs exhibited effects on reducing Tau phosphorylation and A beta aggregation, increasing glucose transporter expression levels, antioxidant enzyme activity, and inhibiting microglial cell activation via the JNK/AKT/GSK3 beta pathway. TG-Res-CS/TPP-NPs also regulated gut microbiota related to inflammation, lipid metabolism, and oxidative stress. This study suggests the potential use of TG-Res-CS/TPP-NPs for Res delivery in the brain.

Synthesis, characterization and evaluation of anti-arthritic and anti-inflammatory potential of curcumin loaded chitosan nanoparticles

Summary: This study successfully formulated curcumin-loaded chitosan and sodium tripolyphosphate nanoparticles to improve the bioavailability of curcumin. The properties and effects of the nanoparticles were characterized and compared. The results showed that curcumin nanoparticles had better anti-inflammatory and anti-arthritic activity compared to curcumin alone. The study concluded that the nanoparticle formulation method can enhance curcumin absorption, reduce the dosing rate, and improve patient compliance.

Aβ-responsive metformin-based supramolecular synergistic nanodrugs for Alzheimer's disease via enhancing microglial Aβ clearance

Summary: In this study, we successfully fabricated metformin-based supramolecular nanodrugs with an A beta-responsive on-demand drug release for synergistic Alzheimer's disease therapy. The nanodrugs exhibited outstanding physiological stability and selectively penetrated the blood-brain barrier to target microglia and neurons. The released drugs, metformin and donepezil, showed a superior synergistic anti-dementia effect by enhancing microglial phagocytosis and A beta clearance.

Summary: The combination of the epigenetic modulator romidepsin and insulin sensitizer metformin improves gene expressions of neurotrophic factors, reduces Alzheimer's disease-associated biochemical and cellular changes, and enhances therapeutic effectiveness and bioavailability through a nanocarrier system. The nanoformulations of romidepsin and metformin significantly reduce stress and increase neurotrophic factors, showing superior neurological effectiveness compared to free drug combinations, ultimately enhancing the synergistic neuroprotective efficacy of the two drugs.

Metformin derived carbon dots: Highly biocompatible fluorescent nanomaterials as mitochondrial targeting and blood-brain barrier penetrating biomarkers

Summary: This study synthesized metformin derived carbon dots (Met-CDs) with low toxicity and excellent biocompatibility, making them ideal candidates for bioimaging studies. Furthermore, Met-CDs were shown to penetrate cell membranes and localize in the nucleus and mitochondria, with potential selective targeting of cancer cell mitochondria. Zebrafish study confirmed Met-CDs crossing the blood-brain barrier without additional ligands.
