A red light-triggered chemical tool for sequence-specific alkylation of G-quadruplex and I-motif DNA

Dual-Color Fluorescence Switch-On Probe for Imaging G-Quadruplex and Double-Stranded DNA in Living Cells

Summary: A tripodal quinone-cyanine dye QCy(MeBT)(3) with one donor and three acceptors was synthesized and demonstrated to exhibit significantly enhanced fluorescence emissions in G4 DNA and double-stranded DNA structures. Docking simulations and viscosity change experiments revealed the reason behind the dual-fluorescence response. Fluorescence microscopy imaging experiments showed the potential of this dye to image G4 DNA and double-stranded DNA in the cell nucleus using near-infrared and red fluorescence emissions, and it could also be used to visualize pyridostatin-induced G4 formation in living cells using near-infrared fluorescence.

Visualization of Deep Tissue G-quadruplexes with a Novel Large Stokes-Shifted Red Fluorescent Benzothiazole Derivative

Structural Basis of Pyridostatin and Its Derivatives Specifically Binding to G-Quadruplexes

Summary: This study reveals the structural basis for the specific recognition of G-quadruplex (G4) by pyridostatin (PDS) and its derivatives. The results demonstrate that the rigid aromatic rings of PDS linked by flexible amide bonds match adaptively with G-tetrad planes, enhancing p-p stacking and achieving specific recognition of G4s. The aliphatic amine side chains of PDS adjust conformation to interact with the phosphate backbone via hydrogen bonding and electrostatic interactions, increasing affinity for G4s.
