Deciphering the catalytic mechanism of superoxide dismutase activity of carbon dot nanozyme

Wenhui Gao et al.

Summary: We report a carbon dot (C-dot) superoxide dismutase (SOD) nanozyme with a catalytic activity comparable to that of natural enzymes. The SOD-like activity of C-dot nanozyme relies on specific chemical modifications and conjugated carbonyl groups for binding and electron transfer. Furthermore, the C-dot SOD nanozymes exhibit intrinsic targeting ability to oxidation-damaged cells and effectively protect neuron cells in the ischemic stroke mouse model.

The Cytokine Network in Colorectal Cancer: Implications for New Treatment Strategies

Heidi Braumueller et al.

Summary: Colorectal cancer (CRC) is a common tumor with limited treatment options. It is a genetic disease with mutations in specific oncogenes and tumor suppressor genes, as well as a multifactorial disease involving environmental factors. The cytokine networks between cancer cells, tumor microenvironment (TME), and microbiome are important in regulating tumor growth, metastasis, and treatment resistance in CRC.

Structural mechanisms for regulation of GSDMB pore-forming activity

Xiu Zhong et al.

Summary: Cytotoxic lymphocyte-derived granzyme A (GZMA) cleaves GSDMB, a gasdermin-family pore-forming protein, to trigger target cell pyroptosis. This study reveals the mechanism of how the Shigella flexneri ubiquitin-ligase virulence factor IpaH7.8 targets both GSDMB and GSDMD. The study also highlights the influence of different splicing isoforms of GSDMB on pyroptotic activity.

NIR-II laser-mediated photo-Fenton-like reaction via plasmonic Cu9S8 for immunotherapy enhancement

Lu An et al.

Summary: Chemodynamic therapy (CDT) is a method used to modulate the tumor microenvironment and extend the benefits of immune checkpoint therapy. A Near-Infrared (NIR)-II laser-mediated photoFenton-like reaction was proposed as an enhancement strategy to enhance the immune response. The experimental results show that this strategy effectively induces immunogenic cell death and dendritic cell maturation.

Real-Time Detection of Hydroxyl Radical Generated at Operating Electrodes via Redox-Active Adduct Formation Using Scanning Electrochemical Microscopy

Jaxiry S. Barroso-Martinez et al.

Summary: The hydroxyl radical (•OH) is a highly attractive reactive oxygen species with potential applications in efficient wastewater treatment and electrosynthesis. However, its short lifetime poses challenges for in situ detection. In this study, we used spin traps and electron spin resonance to detect •OH in solution. We also developed a scanning electrochemical microscopy technique to detect its generation in real time from electrode surfaces. Our findings provide valuable insights for the detection and exploration of •OH in oxidation processes at electrode surfaces.

Application of glutathione depletion in cancer therapy: Enhanced ROS-based therapy, ferroptosis, and chemotherapy

Boyi Niu et al.

Summary: Glutathione (GSH) plays a crucial role in cellular antioxidative system, particularly in cancer cells, where a high level of GSH is essential for scavenging ROS and detoxifying xenobiotics. Depletion of GSH has been shown to enhance the efficacy of various therapies such as ROS-based therapy, ferroptosis, and chemotherapy. This review comprehensively discusses the importance of GSH depletion in cancer therapy and various strategies employed to achieve it.

Constructing Electron Levers in Perovskite Nanocrystals to Regulate the Local Electron Density for Intensive Chemodynamic Therapy

Peiran Zhao et al.

Summary: By regulating the electron density of reaction center atoms, this study has successfully increased the catalytic activity of Fe, reduced the activation energy of the Fenton reaction, and increased the (OH)-O- production rate. This provides a new possibility for highly effective chemodynamic therapy.

Inflammation-related pyroptosis, a novel programmed cell death pathway, and its crosstalk with immune therapy in cancer treatment

Sheng-Kai Hsu et al.

Summary: New types of programmed cell death, such as ferroptosis, necroptosis, and pyroptosis, have emerged in recent years, playing significant roles in cancer progression modulation and being closely related to immune regulation in the tumor microenvironment. Pyroptosis, in particular, has been identified as crucial for host defense and bridging innate and adaptive immunity, showing benefits for cancer immunotherapies.
