MYC overexpression leads to increased chromatin interactions at super-enhancers and MYC binding sites

Yi Xiang See et al.

Summary: The MYC oncogene is frequently dysregulated in various cancer cell types, making it an attractive target for cancer therapy. This study found that MYC overexpression leads to MYC binding at super-enhancers and alters the chromatin landscape of the cancer genome. These findings have implications for understanding and targeting MYC-driven cancers.

JASPAR 2022: the 9th release of the open-access database of transcription factor binding profiles

Jaime A. Castro-Mondragon et al.

Summary: JASPAR is an open-access database containing manually curated, non-redundant transcription factor binding profiles across six taxonomic groups. The latest release introduces new profiles for plants, vertebrates, urochordates, and insects, as well as new tools for TFBS enrichment analysis and data access. The database can be accessed through various platforms, including the JASPAR website, RESTful API, R/Bioconductor data package, and a new Python package pyJASPAR.

Activation of stably silenced genes by recruitment of a synthetic de-methylating module

Wing Fuk Chan et al.

Summary: By coupling the catalytic domain of DNA demethylating enzyme TET1 with transcriptional activators (TETact), the authors have developed an improved activation system that can induce transcription of stably silenced genes. The TETact system is applicable in various cell types and has the ability to reactivate embryonic haemoglobin genes in non-erythroid cells. It is expected that TETact will have significant implications in functional studies, genetic screens, and potential therapeutics.

AP-1 family transcription factors: a diverse family of proteins that regulate varied cellular activities in classical hodgkin lymphoma and ALK plus ALCL

Zuoqiao Wu et al.

Summary: Classical Hodgkin lymphoma (cHL) and anaplastic lymphoma kinase-positive, anaplastic large cell lymphoma (ALK+ ALCL) are B and T cell lymphomas that express CD30 and show aberrant expression of AP-1 family of transcription factors. These proteins play various roles in promoting proliferation, suppressing apoptosis, and evading host immune response. Therapeutic strategies targeting AP-1 transcriptional targets or the signaling pathways they regulate are being explored for the treatment of these lymphomas.

PRC1 drives Polycomb-mediated gene repression by controlling transcription initiation and burst frequency

Paula Dobrinic et al.

Summary: The authors discovered that the Polycomb repressive complex PRC1 functions independently of PRC2 to counteract Pol II binding and regulate transcription initiation by controlling transcriptional burst frequency. This finding provides a mechanistic and conceptual framework for Polycomb-dependent transcriptional control.

Single cell transcriptome atlas of mouse mammary epithelial cells across development

Bhupinder Pal et al.

Summary: The study explored cellular heterogeneity and lineage relationships within the mouse mammary epithelium, revealing different lineage differentiation states during mammary development and the establishment of lineage-specific chromatin during puberty. Distinct cell lineages were identified at specific stages of pregnancy, with a continuum of alveolar-restricted progenitor states along the basal-luminal axis.
