Spin Hyperpolarization in Modern Magnetic Resonance

Order-Unity 13C Nuclear Polarization of [1-13C]Pyruvate in Seconds and the Interplay of Water and SABRE Enhancement

Summary: Signal Amplification By Reversible Exchange in SHield Enabled Alignment Transfer (SABRE-SHEATH) was investigated for rapid hyperpolarization of C-13(1) spins of [1-C-13]pyruvate using parahydrogen. The exchange of pyruvate with an iridium polarization transfer complex can be modulated by temperature and co-ligation conditions. Efficient C-13 polarization was achieved in less than 30 seconds by restricting the chemical exchange of [1-C-13]pyruvate at lower temperatures, with the possibility of fast release for future biomedical applications.

Temperature Cycling Enables Efficient 13C SABRE-SHEATH Hyperpolarization and Imaging of [1-13C]-Pyruvate

Summary: This study introduces a temperature cycling method for SABRE-SHEATH that enables >10% polarization on [1-C-13]-pyruvate, sufficient for successful in vivo experiments. By using SABRE catalyst-bound pyruvate at lower temperatures, approximately 20% polarization is accumulated and released into free pyruvate at elevated temperatures. A kinetic model of differential equations is developed to explain this effect and characterize critical relaxation and buildup parameters, demonstrating the combination of inexpensive hyperpolarization methods with low-cost MRI systems to achieve a broadly available, yet highly sensitive metabolic imaging platform.

Diagnostic imaging is shifting towards functional imaging of pathology for better understanding of individual disease biology and personalized therapies. This study presents a fast and low-cost hyperpolarization method using chemical exchange with parahydrogen, allowing for the cost-effective production of hyperpolarized [1-13C]pyruvate biosensors with great potential for in vivo applications.

Symmetry-based singlet-triplet excitation in solution nuclear magnetic resonance

Summary: Coupled pairs of spin-1/2 nuclei can form one singlet state and three triplet states. The nuclear singlet order, which is the difference between the singlet population and the mean of the triplet populations, is a long-lived state in solution. The PulsePol pulse sequence, based on the theory of symmetry-based recoupling sequences, is an efficient technique for generating long-lived singlet order from magnetization in nuclear magnetic resonance (NMR). This study introduces new pulse sequences for singlet-triplet excitation and conversion in solution NMR and demonstrates schemes to enhance their robustness.
