Experimental evolution of Pseudomonas putida under silver ion versus nanoparticle stress

Feng Dong et al.

Summary: The study found that bacteria displayed significant differences in evolutionary mutations when treated with silver ions or silver nanoparticles, suggesting the presence of nanoparticle-specific effects potentially caused by sustained release of silver ions from nanoparticle dissolution, direct action of nanoparticles on the cell's outer membrane, or more proximate release of ions from cell-surface bound nanoparticles.

Mutations in SilS and CusS/OmpC represent different routes to achieve high level silver ion tolerance in Klebsiella pneumoniae

Charlotte A. Woolley et al.

Summary: This study reveals two possible pathways for the development of increased silver tolerance in Klebsiella pneumoniae, with a higher frequency of mutations in the sil operon. The potential clinical impact is demonstrated by the increased survivability of adapted strains when exposed to silver-containing dressings, making infections with these strains more difficult to treat and further limiting therapeutic options.

Structures of the Type IX Secretion/Gliding Motility Motor from across the Phylum Bacteroidetes

Rory Hennell James et al.

Summary: Gliding motility and protein secretion through the type IX secretion system (T9SS) are two specific features of the Bacteroidetes. These processes are energized by the GldLM motor complex, which converts the proton motive force into mechanical work. The structures of the GldLM motor complex from different Bacteroidetes species reveal a conserved architecture, but with species-specific differences at the N terminus of GldL. This study enhances our understanding of bacterial ion-driven motors and their role in T9SS and gliding motility.

Characterization of Silver Resistance and Coexistence of sil Operon with Antibiotic Resistance Genes Among Gram-Negative Pathogens Isolated from Wound Samples by Using Whole-Genome Sequencing

Haichen Wang et al.

Summary: This study investigated the silver resistance mechanisms and coexistence of antibiotic resistance genes with the sil operon among gram-negative pathogens isolated from wound samples. The results showed a prevalent cryptic silver resistance among Enterobacteriaceae with the sil operon or in combination with the cus operon and major porin loss, along with the presence of antibiotic resistance genes.

Poly-Gamma-Glutamic Acid Secretion Protects Bacillus subtilis from Zinc and Copper Intoxication

Reina Deol et al.

Summary: Zinc and copper are essential micronutrients that are toxic to bacteria at elevated concentrations. Bacteria have evolved defense mechanisms, including the production of extracellular polymeric substance poly-gamma-glutamate (gamma-PGA) to combat zinc and copper excess. This study provides new insight into metal resistance mechanisms and regulation of bacterial gamma-PGA biosynthesis.

Mutagenesis and Resistance Development of Bacteria Challenged by Silver Nanoparticles

Kun Wu et al.

Summary: Silver nanoparticles (AgNPs) have broad-spectrum antimicrobial properties, but their mutagenic effects and resistance mechanisms in bacteria are still controversial. This study found that AgNPs do not accelerate resistance mutation generation by increasing the mutation rate, and they also inhibit quorum sensing in bacteria. However, bacteria can still develop resistance through two-component regulatory systems.

Osmotic stress and vesiculation as key mechanisms controlling bacterial sensitivity and resistance to TiO2 nanoparticles

Christophe Pagnout et al.

Summary: This study investigates the response of lipopolysaccharide-truncated Escherichia coli mutants to TiO2 nanoparticles, showing that different lipopolysaccharide surface phenotypes can affect bacterial sensitivity to the nanoparticles. TiO2 nanoparticles strip the cell membrane, leading to osmotic stress, oxidative stress, and the formation of membrane vesicles, which can act as electrostatic baits for the nanoparticles, highlighting an antagonistic bacterial response to nanoparticle toxicity.

Additionally, the research demonstrates that the intensity of side-scatter light parameter increases with TiO2 and ZnO NPs in a dose-dependent manner, indicating NP uptake by both strains.

Role of bacterial motility in differential resistance mechanisms of silver nanoparticles and silver ions

The study of bacterial resistance to AgNPs is still in its early stages, with limited understanding of the underlying mechanisms. The resistance to subinhibitory concentrations of AgNPs in Escherichia coli K-12 MG1655 strain was found to be associated with a cusS mutation, potentially enhancing silver ion efflux and suggesting a role for flagellum-based motility in the resistance mechanism.
