Uncovering the riddle of femininity in osteoarthritis: a systematic review and meta-analysis of menopausal animal models and mathematical modeling of estrogen treatment

Summary: This study aims to summarize the current understanding of the effects of menopause on osteoarthritis (OA) in animal models and investigate the impact of dosage and timing of estrogen treatment on cartilage degeneration. The results show that post-menopausal animals have worse cartilage outcomes compared to non-menopausal animals, and early initiation and higher doses of estrogen treatment may improve cartilage health.

Age-related matrix stiffening epigenetically regulates α-Klotho expression and compromises chondrocyte integrity

Summary: Matrix stiffening is a key characteristic of aged tissues. In this study, the authors found that an aged (stiff) matrix causes epigenetic repression of the gene encoding alpha-Klotho, a longevity factor, leading to chondrocyte dysfunction, which is a major cause of osteoarthritis. Exposing aged chondrocytes to a soft matrix restores a more youthful phenotype and enhances cartilage integrity. These findings highlight the importance of age-related biophysical alterations in extracellular matrix and have implications for the broader field of aging research.

Sex-dependent variation in cartilage adaptation: from degeneration to regeneration

Summary: Despite recognition of sex-based differences in cartilage biology, the implications for study design and treatment efficacy remain unclear. This paper summarizes current findings on sex-dependent variation in knee anatomy, sex hormones' effect on cartilage, and cartilaginous degeneration and regeneration, with a focus on stem cell therapies. The contribution of both stem cell donor and recipient sex to sex-related differences in treatment efficacy is highlighted. More research is needed to optimize strategies for cartilage regeneration, taking sex into account for more effective and personalized treatments.

Hallmarks of aging: An expanding universe

Summary: Aging is driven by hallmarks that manifest with age, accelerate aging when accentuated experimentally, and can be decelerated, stopped, or reversed with therapeutic interventions. The twelve proposed hallmarks of aging include genomic instability, telomere attrition, epigenetic alterations, loss of proteostasis, disabled macroautophagy, deregulated nutrient-sensing, mitochondrial dysfunction, cellular senescence, stem cell exhaustion, altered intercellular communication, chronic inflammation, and dysbiosis. These hallmarks are interconnected with each other and with the recently proposed hallmarks of health.

This review examines the state of preclinical aging research in terms of sex differences and suggests strategies for improving adherence to the mandate. Considering sex as a biological variable and including both males and females in aging biology research is crucial for improving health outcomes for all.
