MIBlood-EV: Minimal information to enhance the quality and reproducibility of blood extracellular vesicle research

Benchmarking blood collection tubes and processing intervals for extracellular vesicle performance metrics

Summary: The analysis of extracellular vesicles (EV) in blood samples is being extensively studied, as it has the potential to provide clinically meaningful biomarkers for health and disease. This study investigated the impact of different blood collection tubes and processing intervals on EV characteristics in blood samples. The results showed that the choice of blood collection tube and processing interval significantly affected various metrics related to blood sample quality, ex-vivo generation of EV from blood cells, EV recovery, and molecular signatures associated with EV. These findings provide valuable guidance for selecting optimal conditions for EV analysis and contribute to standardizing EV studies.

Platelet removal from human blood plasma improves detection of extracellular vesicle-associated miRNA

Summary: This study investigated whether the analysis of EV-miRNA is affected by the presence of platelets and ery-ghosts in human plasma. The results showed that filtration of plasma can remove platelets and ery-ghosts, improving the comparability and reproducibility of EV-miRNA studies. The authors recommend measuring and reporting the plasma concentration of platelets for EV-miRNA studies, and filtering plasma before downstream analyses or storage in biobanks.

A Simple and Cost-Effective Method for Measuring Hemolysis in Biobank Serum Specimens

Summary: In this study, a simple and cost-effective NanoDrop-based method was developed for measuring hemolysis levels in biobank samples. Centrifugation, instead of temperature, was found to be a crucial step in reducing interference from sample turbidity. Long-term stored samples showed a strong effect of centrifugation on turbidity, and treatment at 20,000 g for 15 minutes at 19 degrees C reduced turbidity up to 50%. Fresh samples experienced a slight increase in optical density after a single freeze-thaw cycle, which was corrected by subsequent centrifugation.

Robust sequential biophysical fractionation of blood plasma to study variations in the biomolecular landscape of systemically circulating extracellular vesicles across clinical conditions

Summary: This study fractionated blood plasma using SEC and OptiPrep density gradient centrifugation to investigate clinical and time-dependent variations in the biomolecular landscape of systemically circulating EV. Results showed that EV carry context-dependent and/or time-dependent protein and small RNA compositions, with differential analysis providing a catalogue of proteins not associated with EV. Overall, blood plasma fractionation can advance the biological understanding and biomarker development of systemically circulating EV.
