Progress in Antimelanoma Research of Natural Triterpenoids and Their Derivatives: Mechanisms of Action, Bioavailability Enhancement and Structure Modifications

The Antimelanoma Biological Assessment of Triterpenic Acid Functionalized Gold Nanoparticles

Summary: Gold nanoparticle formulation is a promising strategy for enhancing the bioavailability and therapeutic potential of high-lipophilic biologically active compounds. In this study, three triterpen-functionalized gold nanoparticles were synthesized and evaluated for their biological antimelanoma activity. The conjugates exhibited dose-dependent cytotoxic effects on melanoma cells, but no significant toxicity on normal keratinocytes. The functionalized gold nanoparticles showed higher cytotoxicity compared to their parent compounds and induced morphological changes consistent with apoptosis in melanoma cells.

Dammarane-Type Triterpenoid from the Stem Bark of Aglaia elliptica (Meliaceae) and Its Cytotoxic Activities

Summary: In this study, two new dammarane-type triterpenoid fatty acid ester derivatives were isolated from the stem bark of Aglaia elliptica. The structures of these compounds were determined using various spectroscopic methods and comparison with previous literature. The cytotoxicity of these compounds, along with their synthetic analogs, was evaluated against breast cancer and melanoma cell lines. The results showed that one compound exhibited strong activity against both cell lines, while the other compounds had weak activity, indicating the significance of the presence of fatty acids in the cytotoxicity of dammarane-type triterpenoids.

Summary: In the search for potential medicinal substances for oncology, new compounds combining the betulin molecule and the indole system were synthesized. The structure of the obtained derivatives was confirmed and their anticancer activity was evaluated. The results indicated that one derivative showed promising cytotoxic effect on breast cancer cells, making it a potential starting point for further research in breast cancer therapy or synthesis of new derivatives.

Further pharmacological experiments revealed that compound 8a could induce morphological changes, cell cycle arrest, and apoptosis in HepG2 cells, suggesting its potential as a rapid strategy for the discovery of antiproliferative agents.

Molecular Structure, In Vitro Anticancer Study and Molecular Docking of New Phosphate Derivatives of Betulin

Summary: A series of 30-diethylphosphate derivatives of betulin were synthesized and evaluated for their cytotoxic activity against human cancer cell lines. Compounds 7a and 7b showed the highest activity against C-32 and SNB-19 cell lines, with IC50 values of 2.15 and 0.91 mu M for 7a, and 0.76 and 0.8 mu M for 7b. The most active compounds were further studied for their effects on apoptosis markers and docked to the active site of the EGFR protein, showing significant interactions and stable complexes.
