Switching Nitrofurantoin Polymorphic Outcome in Solvent-Mediated Phase Transformation and Crystallization Using Solvent and Additives

Summary: Several additives and solvents can switch the polymorphic outcome of solvent-mediated phase transformation experiments and crystallization of nitrofurantoin. The selection of alcohols or the use of crystallization additives leads to the formation of polymorph α, while most solvents result in polymorph β. This can be attributed to the reversed apparent relative solubility of nitrofurantoin polymorphs in different solvents or in the presence of additives.

Crystallization of Form II Paracetamol with the Assistance of Carboxylic Acids toward Batch and Continuous Processes

Summary: Form II paracetamol, with improved compressibility, has attracted researchers' attention. This study focused on the selective polymorphic formation of paracetamol using cooling crystallization with four types of additives. The results showed that the addition of more additives increased the probability of forming Form II paracetamol. By seeding the paracetamol solution with Form II paracetamol and fumaric acid crystals, Form II paracetamol could be induced, while the other three carboxylic acids were not effective. The study also discovered a new solution complex of paracetamol-oxalic acid, responsible for the selective nucleation of Form II paracetamol in the oxalic acid solution. Additionally, the range of supersaturation for nucleating Form II paracetamol was extended with the assistance of oxalic acid or fumaric acid. In large-scale crystallization, Form II paracetamol was successfully produced using continuous crystallization in a fumaric acid aqueous solution.

Concomitant Polymorphism and Nucleation Control of DL-Methionine Through Antisolvent Crystallization

Summary: In this study, researchers discovered the concomitant polymorphism of the sulfur-containing essential amino acid DL-methionine and successfully separated the alpha- and beta-polymorphic forms through antisolvent crystallization with ethanol. The addition of varying volume fractions of ethanol into the solution affected the solubility of DL-methionine and led to different levels of supersaturation, resulting in the formation of stable beta-polymorphs, mixture of alpha- and beta-polymorphs, and metastable alpha-polymorphs. The distinct needle-shaped alpha-polymorphs and platy-shaped beta-polymorphs were confirmed through optical microscopic and X-ray diffraction analyses.

Switching polymorph stabilities with impurities provides a thermodynamic route to benzamide form III
