Taking Charge: Metal Ions Accelerate Amyloid Aggregation in Sequence Variants of ?-Synuclein

Summary: Alpha-synuclein is a protein with high structural diversity. It undergoes conformational adaptation in different cellular environments. Divalent metal ions, such as Ca2+, Mn2+, and Zn2+, known to bind to the C-terminal region of alpha-synuclein, are abundant in synaptic terminals where alpha-synuclein is located. In this study, native nanoelectrospray ionization ion mobility-mass spectrometry was used to investigate the effects of divalent metal ions on the conformation and amyloid formation behavior of alpha-synuclein variants. The results demonstrate that specific intramolecular interactions govern the conformational ensemble of alpha-synuclein and influence its amyloidogenic behavior.

However, studying the metastable oligomers and their interactions with potential therapeutics is challenging, hindering the development of disease-modifying therapies. This article analyzes the utilization of ion-mobility spectroscopy-MS (IM-MS) and electron-capture dissociation (ECD) MS/MS for studying oligomerization and conformation of amyloidogenic proteins, as well as their interaction with drug candidates.

The results provide insights into the molecular mechanisms of how CLR01 affects the stability of SOD1 and suggest it as a potential target for the development of drugs against neurodegenerative diseases.
