A Mitochondria-Targeted Ferroptosis Inducer Activated by Glutathione-Responsive Imaging and Depletion for Triple Negative Breast Cancer Theranostics

Hongbo Gan et al.

Summary: A theranostic ferroptosis inducer (IR780-SPhF) targeting mitochondria was developed for triple-negative breast cancer (TNBC) treatment. It reacts with GSH to deplete mitochondrial GSH and disrupt redox balance, while also exhibiting GSH-responsive near-infrared fluorescence emission and photoacoustic imaging characteristics for real-time monitoring and diagnosis. Both in vitro and in vivo results showed that IR780-SPhF possessed potent anticancer effect, surpassing the classic drug cyclophosphamide commonly used for TNBC patients.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

lncRNA LUCAT1/ELAVL1/LIN28B/SOX2 Positive Feedback Loop Promotes Cell Stemness in Triple-Negative Breast Cancer

Li Xia et al.

Summary: This study investigated the role of long noncoding RNA LUCAT1 in triple-negative breast cancer (TNBC) and its potential regulatory mechanism. The results showed that LUCAT1 was significantly overexpressed and could recruit ELAVL1 protein to stabilize LIN28B mRNA, further regulating the expression of SOX2, forming a positive feedback loop that promoted cell stemness.

Non-coding RNAs in ferroptotic cancer cell death pathway: meet the new masters

Mehdi Rabiee Valashedi et al.

Summary: Cancer cells' resistance to ferroptosis, a new form of regulated cell death, is a major challenge in cancer therapy. Non-coding RNAs have been found to play a role in regulating ferroptosis, and targeting these RNAs could potentially overcome cancer drug resistance. Gene therapy, including gene-editing technology, may aid in developing ncRNA-based ferroptosis targeting as a novel therapeutic strategy for cancer treatment.

Long non-coding RNA LUCAT1 regulates the RAS pathway to promote the proliferation and invasion of malignant glioma cells through ABCB1

Xia Wu et al.

Summary: This study investigated the role of lncRNA LUCAT1 in the occurrence and development of glioma, as well as its regulatory mechanism. The results demonstrated that LUCAT1 can affect the biological behaviors of glioma cells by regulating ABCB1 and promoting the activation of the RAS signaling pathway, providing a new drug target and therapeutic approach for gene therapy of glioma.

Prediction of Prognosis and Molecular Mechanism of Ferroptosis in Hepatocellular Carcinoma Based on Bioinformatics Methods

Yuanpeng Xiong et al.

Summary: This study identified 93 lncRNAs associated with ferroptosis that are of prognostic value in HCC patients. A five-lncRNA signature, consisting of AC015908.3, LINC01138, AC009283.1, Z83851.1, and LUCAT1, was established to effectively predict the prognosis of HCC patients. The signature was associated with tumorigenesis and immune-related pathways, and could be used to evaluate the clinical efficacy of chemotherapy and immunotherapy.

Epidemiology, Screening, and Prevention of Bladder Cancer

Niyati Lobo et al.

Summary: Bladder cancer is the tenth most common cancer worldwide, with a higher incidence in men than women. Tobacco smoking is the primary risk factor, accounting for approximately 50% of cases. Routine screening is not recommended, but targeted screening for high-risk individuals may reduce mortality. Smoking cessation is the most important prevention measure.

Summary: This study aimed to compare intravesical maintenance Bacillus Calmette-Guerin (mBCG) and radical cystectomy (RC) in high-grade nonmuscle invasive bladder cancer (HRNMIBC), but recruitment was challenging. Results showed around 10% of patients have lethal disease suitable for radical treatment, while many are candidates for bladder preservation with maintained quality of life.
