Summary: The ocular safety and efficacy of tonabersat were evaluated in an inflammatory NOD mouse model of diabetic retinopathy (DR). The results showed that tonabersat significantly reduced inflammation and disease progression, suggesting that it may be an effective treatment for DR.

Summary: This study shows that therapeutic hypothermia can prevent lesions after cerebral ischemia, but it only partially suppresses abnormal morphology and phenotype of microglia. These findings support the hypothesis that persistent upregulation of injurious microglial activity may contribute to partial neuroprotection after hypothermia, and that immunomodulation may be an important therapeutic target.

Connexins, Pannexins and Gap Junctions in Perinatal Brain Injury

Summary: Perinatal brain injury, caused by hypoxia-ischemia and/or infection/inflammation, is a major cause of disability. Despite the effectiveness of therapeutic hypothermia, additional interventions are needed as nearly half of the infants still have adverse outcomes. Recent studies have shown that brain injury spreads over time from the initial site to previously unaffected regions, partly due to the opening of connexin hemichannels and pannexin channels. Blockade of these channels has demonstrated neuroprotective effects in animal models, suggesting their potential as targeted interventions in combination or separate from therapeutic hypothermia.

Summary: Recent research has shown that connexin43 hemichannel blockers can inhibit activation of the inflammasome pathway in diabetic retinopathy. The development of an ex vivo human retinal organotypic culture model provides a more accurate representation of the inflammatory process in diabetic retinopathy, supporting the potential use of connexin43 hemichannel blockers as new therapies for the disease.
