Recent advances in cellular and molecular mechanisms of IgE-mediated food allergy

Jesus F. Crespo et al.

Summary: Food allergy is a growing public health issue, and recent research has shed light on the molecular and cellular mechanisms behind IgE-mediated food allergy. These discoveries have highlighted the important roles of specific cell types, such as T follicular helper cells, in the production of high-affinity IgE. Additionally, it has been found that not only mast cells and basophils, but also other cell types like neutrophils and macrophages, contribute to food anaphylaxis. Understanding the mechanisms of sensitization through organs like the skin is crucial for comprehending the dual exposure hypothesis and uncovering new approaches to anaphylaxis pathophysiology.

Allergenicity of peanut allergens and its dependence on the structure

Qin Geng et al.

Summary: Food allergies, particularly peanut allergies, are a global concern in food safety. This review focuses on the mechanisms, management, and structures of allergens associated with peanut allergies. Epitope localization methods are summarized to aid in the discovery of allergenicity-dependent structures. Efforts to reduce the allergenicity of peanuts include analysis of hypoallergens produced during processing. Identifying and modifying conformational epitopes will lead to improved strategies for managing and preventing peanut allergies, with a potential for application in other food allergies as well.

How Do Pollen Allergens Sensitize?

Svetlana V. Guryanova et al.

Summary: Plant pollen is a major allergen causing allergic diseases such as allergic rhinitis and asthma. The interaction between allergens and molecular factors, as well as their interaction with lipids and other hydrophobic molecules, play important roles in pollen sensitization.

The global burden of illness of peanut allergy: A comprehensive literature review

Jay A. Lieberman et al.

Summary: Peanut allergy (PA) affects around 2% of the general population in Western countries, leading to significant burden of self-management for patients and their families/caregivers. PA is associated with higher rates of accidental exposure, severe reactions, and potential fatal anaphylaxis compared to other food allergies, resulting in considerable healthcare utilization and economic costs.

Update on food allergy

Rachel L. Peters et al.

Summary: Food allergy is a growing public health issue in urbanized areas, with research into risk factors and immune mechanisms playing a key role in prevention and treatment. Introducing allergenic foods in the diet may help prevent the development of food allergy, while improved diagnosis and management are essential for patient care.

Characteristics of Peanut Allergy Diagnosis in a US Health Care Claims Database (2011-2017)

Todd A. Mahr et al.

Summary: This study estimated the annual incidence and prevalence of peanut allergy in children in the United States, as well as assessed the rates of severe reactions and associated health care utilization. The results suggest that the burden of peanut allergy may be considerable, with high rates of severe reactions and emergency department visits.

Prevalence and characteristics of peanut allergy in US adults

Christopher Warren et al.

Summary: The prevalence of self-reported peanut allergy among US adults is 2.9%, with over 17% of cases being adult-onset. Adults with childhood-onset PA are more likely to have received a physician diagnosis, have an epinephrine prescription, and use an epinephrine autoinjector compared to those with adult-onset PA.

Pathogenesis of IgE-mediated food allergy and implications for future immunotherapeutics

Nicole Ramsey et al.

Summary: Research on the immune basis and treatment of food allergy is rapidly advancing, with a focus on local tissue factors and cytokines in the development and treatment of food allergy. The role of IL-13 and the targeting of IL-4 and IL-13 are significant in the treatment of food allergy, while low specific IgE and elevated specific IgG4 are predictive of treatment outcomes.

Does the epithelial barrier hypothesis explain the increase in allergy, autoimmunity and other chronic conditions?

Cezmi A. Akdis

Summary: The increase in allergic, autoimmune, and chronic diseases may be related to epithelial barrier-damaging agents associated with industrialization, urbanization, and modern life. These diseases are more prevalent in industrialized countries and are on the rise in developing countries, affecting over one billion people globally.
