Graded BMP signaling within intestinal crypt architecture directs self-organization of the Wnt-secreting stem cell niche

Summary: Signals from the surrounding niche regulate the behavior of intestinal stem cells (ISCs) by driving their proliferation and suppressing their differentiation. Stromal cells, such as trophocytes, provide essential factors for ISC maintenance and their expression gradually changes spatially and molecularly. The graded expression of niche factors is influenced by the distance from BMP-secreting myofibroblast aggregates, leading to the establishment of a self-organized and polar ISC niche.

Summary: This study reveals that gliomas can influence neural circuits and cognition through bidirectional interactions with neurons. Gliomas remodel functional neural circuitry and promote tumor progression while impairing cognition. The degree of functional connectivity between glioblastoma and the normal brain negatively affects patient survival and performance in language tasks.

Summary: Epigenetic abnormalities, such as G9a depletion, can inhibit the migration and invasion abilities of breast cancer cells, and enhance their response to ionizing radiation. This study identifies BMP5 as a downstream target gene of G9a, and finds that increased BMP5 expression leads to the phosphorylation of Smad proteins, thereby reducing breast cancer cell migration and invasion.

BMP gradient along the intestinal villus axis controls zonated enterocyte and goblet cell states

Summary: By studying human intestinal organoids and mouse models, it was found that the bone morphogenetic protein (BMP) signal drives the zonation of intestinal absorptive cells and goblet cells. These findings suggest that the regulation of BMP signal strength can control carbohydrate and lipid uptake in enterocytes and antimicrobial response through goblet cells.

BMP2 promotes lung adenocarcinoma metastasis through BMP receptor 2-mediated SMAD1/5 activation

Summary: This study revealed the overexpression of BMP2 in lung adenocarcinoma patients with lymph node metastasis. In an in vivo mouse model, BMP2 was shown to promote lung adenocarcinoma metastasis. The activation of the SMAD1/5/8 signaling pathway was found to be involved in BMP2-mediated cell migration. Therefore, targeting the BMP2 signaling pathway could be a potential therapeutic strategy for treating metastatic lung adenocarcinoma.

Summary: Xi and colleagues demonstrate that BMP signaling has tumor-suppressive effects in ACVR1 wild-type diffuse intrinsic pontine glioma by modulating stemness via chromatin regulation of CXXC5. They also show the therapeutic potential of this pathway using in vivo mouse models.

CREB signaling activity correlates with differentiation and survival in medulloblastoma

Summary: The study found that phosphorylation of the transcription factor CREB correlates strongly with survival in medulloblastoma patients and differentiation phenotype. During normal cerebellar development, phosphorylated CREB is selectively expressed in differentiating cerebellar granule neuron progenitor cells. Inducing CREB activation via PKA-mediated signaling enhances sensitivity of medulloblastoma cells to chemotherapy.
