Mitochondrial quality control mechanisms as therapeutic targets in doxorubicin-induced cardiotoxicity

Summary: Doxorubicin (DOX) is effective in treating solid tumors and hematologic malignancies but its clinical application is limited by its cardiotoxicity. Defective mitochondrial quality control has been found to play a major role in the development of DOX-induced cardiomyopathy. Targeting these quality control mechanisms has emerged as a potential therapeutic strategy to attenuate DOX cardiotoxicity. However, clinical translation is challenging due to obscure mechanisms of action and potential adverse effects.

Summary: This study aimed to determine the therapeutic efficiency and action mechanism of bardoxolone methyl (BARD) in myocardial ischemia/reperfusion (I/R) injury. The results showed that BARD could reduce cardiac injuries, decrease cardiomyocyte apoptosis, and inhibit oxidative stress. Mechanistically, BARD treatment significantly activated the Nrf2/HO-1 pathway.

Curcumin Improves Diabetic Cardiomyopathy by Inhibiting Pyroptosis through AKT/Nrf2/ARE Pathway

Summary: This study investigates the effects of curcumin on diabetic cardiomyopathy by regulating the AKT pathway and promoting the transfer of Nrf2 into the nucleus. Curcumin was found to inhibit cell pyroptosis by reducing reactive oxygen species accumulation and mitochondrial damage in diabetic myocardium. The protective effects of curcumin were diminished when the Nrf2 pathway was blocked. These findings suggest that curcumin has potential therapeutic effects in treating diabetic cardiomyopathy.

Pharmacological senolysis reduces doxorubicin-induced cardiotoxicity and improves cardiac function in mice

Summary: Many anticancer drugs can cause premature aging in healthy tissues, resulting in accelerated aging processes and adverse effects in patients. Cardiotoxicity is a major limiting factor in the use of doxorubicin as a cancer treatment, leading to long-term morbidity and mortality. In this study, it was demonstrated that induction of senescence in the heart contributes to impaired cardiac function in mice treated with doxorubicin. Elimination of senescent cells using the senolytic Navitoclax significantly reduced senescence and cardiotoxicity markers, and restored cardiac function in mice. These results suggest the potential clinical use of senolytic therapies to alleviate cardiotoxicities induced by chemotherapy.
