AaMYB108 is the core factor integrating light and jasmonic acid signaling to regulate artemisinin biosynthesis in Artemisia annua

Hang Liu et al.

Summary: In this study, a transcription factor named AaMYB108 was identified as a positive regulator of artemisinin biosynthesis in A. annua. AaMYB108 promotes artemisinin biosynthesis by interacting with a previously characterized positive regulator, AaGSW1. Additionally, AaMYB108 interacts with AaCOP1 and AaJAZ8, and its function is influenced by these proteins. Through experiments with transgenic plants, it was found that the promotion of artemisinin by light and JA depends on the presence of AaMYB108. Overall, this study reveals the molecular mechanism of JA regulating artemisinin biosynthesis depending on light and provides new insights into the integration of light and phytohormone signaling in plants.

AaSEPALLATA1 integrates jasmonate and light-regulated glandular secretory trichome initiation in Artemisia annua

Tian-Tian Chen et al.

Summary: In this study, a MADS-box transcription factor AaSEP1, induced by jasmonate and light signaling, was identified as a positive regulator of glandular secretory trichome (GST) initiation in Artemisia annua. AaSEP1 enhanced the function of AaHD1 activation and interacted with AaMYB16 and AaJAZ8, contributing to GST initiation mediated by the jasmonate signaling pathway. AaSEP1 also interacted with AaCOP1, a major repressor of light signaling. This study provides important insights into the regulatory mechanism of GST initiation in A. annua.

Summary: Exploring the genetic network of glandular trichomes and manipulating genes relevant to secondary metabolite biosynthesis are of great importance and value. Artemisinin, a key antimalarial drug ingredient, is synthesized and stored in glandular secretory trichomes (GSTs) in Artemisia annua. WIN/SHN proteins, a clade of AP2/ERF family, are known as regulators for cuticle biosynthesis. However, their function in glandular trichome development is less unknown.

AaWRKY17, a positive regulator of artemisinin biosynthesis, is involved in resistance to Pseudomonas syringae in Artemisia annua

Tiantian Chen et al.

Summary: In this study, a novel WRKY transcription factor AaWRKY17 was identified in Artemisia annua, which could increase artemisinin content and resistance to Pseudomonas syringae. AaWRKY17 directly bound to the promoter region of ADS gene and promoted its expression, leading to increased levels of defense marker genes. Overexpression of AaWRKY17 in A. annua resulted in enhanced pathogen tolerance and artemisinin content. This suggests that AaWRKY17 could be used in transgenic breeding to improve artemisinin production and pathogen tolerance in A. annua.
