Examination of two different proteasome inhibitors in reactivating mutant human cystathionine β-synthase in mice

Sapna Gupta et al.

Summary: Through experiments, it has been proven that proteasome inhibitors can restore the function of mutant CBS protein in mouse models. Two FDA-approved proteasome inhibitors, carfilzomib and bortezomib, were effective in inducing chaperone proteins and inhibiting proteasome function. However, bortezomib showed a better performance in restoring mutant CBS function.

Recent therapeutic approaches to cystathionine beta-synthase-deficient homocystinuria

Tomas Majtan et al.

Summary: Homocystinuria, a common metabolic disorder, is characterized by increased homocysteine levels, leading to various clinical complications. Current treatment primarily involves dietary interventions, but alternative therapies such as enzyme and gene therapies have been explored.

How to fix a broken protein: restoring function to mutant human cystathionine β-synthase

Warren D. Kruger

Summary: Inherited errors of metabolism are a group of recessive genetic diseases caused by missense mutations, leading to cellular metabolic disorders. Deficiency of cystathionine beta-synthase is an example, resulting in elevated blood homocysteine levels and various symptoms. Modulation of the intracellular protein folding environment may offer a potential therapeutic strategy for treating this condition.

Summary: The family of HSPBs plays a crucial role in maintaining proteostasis and cellular homeostasis in response to stress and apoptosis. They prevent protein aggregation, promote refolding, and can be associated with various diseases when mutations occur. High expression levels of HSPBs are found in cardiac muscle, skeletal muscle, and neurons, and mutations in specific HSPBs can lead to cardiomyopathies, myopathies, and cataract.

Cystathionine β-synthase deficiency in the E-HOD registry-part I: pyridoxine responsiveness as a determinant of biochemical and clinical phenotype at diagnosis

Viktor Kozich et al.

Summary: The clinical spectrum of CBS deficiency varies widely, with different levels of pyridoxine responsiveness associated with varying disease manifestations and outcomes. Age at diagnosis, age of onset, and treatment effectiveness are influenced by pyridoxine responsiveness. Standardized responsiveness tests and critical evaluation of past assessments are essential for preventing long-term complications in CBS deficiency patients.
