Cordyceps inhibits ceramide biosynthesis and improves insulin resistance and hepatic steatosis

Ying Li et al.

Summary: Ectopic ceramide accumulation in insulin-responsive tissues contributes to the development of obesity and impairs insulin sensitivity. Pharmacological inhibition of serine palmitoyl transferase (SPT), the first enzyme essential for ceramide biosynthesis, using myriocin in rodents reduces body weight and improves insulin sensitivity. Commercially available Cordyceps containing myriocin can reduce ceramide accrual, increase energy expenditure, prevent diet-induced obesity, improve glucose homeostasis, and resolve hepatic steatosis.

Significance of Brain Glucose Hypometabolism, Altered Insulin Signal Transduction, and Insulin Resistance in Several Neurological Diseases

Enrique Blazquez et al.

Summary: Several neurological diseases share similar pathological alterations, including neuroinflammation and altered brain glucose metabolism. Insulin and brain glucose metabolism abnormalities are considered as key pathological substrates for these diseases and may serve as potential therapeutic targets.

Summary: This article discusses the impact of Alzheimer's disease on public health, including its incidence, prevalence, mortality and morbidity rates, healthcare costs and caregivers' burden. It also addresses the perspectives of consumers and primary care physicians on mild cognitive impairment (MCI) and the need for improved awareness, diagnosis, and treatment. The article emphasizes the increasing number of individuals living with Alzheimer's and the challenges faced by caregivers and healthcare providers, particularly during the COVID-19 pandemic.

APOE4 disrupts intracellular lipid homeostasis in human iPSC-derived glia

Grzegorz Sienski et al.

Summary: The APOE gene's E4 allele is a genetic risk factor for cardiovascular diseases and Alzheimer's disease, but its mechanism of action is poorly understood. A study found that APOE4 disrupted cellular lipidomes, leading to increased unsaturation of fatty acids and intracellular lipid droplet accumulation. Supplementation with choline restored the cellular lipidome in APOE4-expressing cells, suggesting that manipulating lipid metabolism could be a therapeutic approach for individuals with the APOE4 allele.
