Acylcarnitines: Nomenclature, Biomarkers, Therapeutic Potential, Drug Targets, and Clinical Trials

Maija Dambrova et al.

Summary: Acylcarnitines are fatty acid metabolites that play important roles in cellular energy metabolism and have potential as biomarkers and pharmaceutical agents for various diseases. This comprehensive review provides detailed information on the identity, nomenclature, classification, biochemistry, pathophysiology, and clinical trials of acylcarnitines. It also discusses their use as dietary supplements and the emerging opportunities for using them as biomarkers and interventions in different indications. The review lays a solid foundation for further understanding the physiological roles of acylcarnitines and the identification of new drug targets.

The Metabolome and the Gut Microbiota for the Prediction of Necrotizing Enterocolitis and Spontaneous Intestinal Perforation: A Systematic Review

Laura Moschino et al.

Summary: This systematic review examines the use of untargeted metabolomics and gut microbiota analysis in evaluating the differences between neonates affected by NEC and healthy controls. The review finds that an early urinary alanine/histidine ratio >4 may serve as a predictive biomarker for NEC. Additionally, NEC specimens show increased relative abundance of Proteobacteria at phylum level and a lack or decreased abundance of Bifidobacterium at genus level. The study suggests that untargeted metabolomics and gut microbiota analysis can shed light on the pathophysiology of NEC and help identify potential biomarkers.

Mitochondrial dysfunction in inflammatory bowel disease alters intestinal epithelial metabolism of hepatic acylcarnitines

Sarah A. Smith et al.

Summary: This study demonstrates that the intestinal epithelium can oxidize long-chain fatty acids and consume acylcarnitines from bile through absorption at the apical membrane, leading to mitochondrial oxidation. Intestinal inflammation can cause mitochondrial dysfunction in the apical domain of the surface epithelium, reducing fatty acid consumption and resulting in elevated fecal acylcarnitine levels in colitis and inflammatory bowel disease patients. The findings highlight the importance of both the gut microbiota and liver in delivering energy substrates for mitochondrial metabolism in the intestinal epithelium.

Voluntary wheel running is capable of improving cognitive function only in the young but not the middle-aged male APPSwe/PS1De9 mice

Guiping Wang et al.

Summary: The study found that voluntary wheel running improved cognitive function in young APP/PS1 mice through various mechanisms, such as reducing tau phosphorylation and neuro-inflammation, and increasing key synaptic plasticity-related proteins. For middle-aged APP/PS1 mice, voluntary exercise reduced protein expression in certain brain regions, but did not significantly affect circulating metabolites.

An intercrypt subpopulation of goblet cells is essential for colonic mucus barrier function

Elisabeth E. L. Nystrom et al.

Summary: Intestinal goblet cells are not homogeneous, with several distinct subtypes forming two differentiation trajectories. Intercrypt goblet cells located at the colonic luminal surface produce mucus with different properties from crypt-residing goblet cells. Defective icGCs in mice lead to increased sensitivity to chemically induced colitis and spontaneous colitis with age, while alterations in mucus and reduced numbers of icGCs are observed in patients with ulcerative colitis, highlighting the importance of icGCs in maintaining functional epithelial protection.
