Assessment of the Acute Effects of 2C-B vs. Psilocybin on Subjective Experience, Mood, and Cognition

Pablo Mallaroni et al.

Summary: 2C-B is a hallucinogenic phenethylamine derived from mescaline, and it has been found to produce subjective and emotional effects similar to other classical psychedelics and entactogens. This study aimed to assess the acute effects of 2C-B compared to psilocybin and placebo. The results showed that 2C-B induced psychedelic alterations of consciousness, with dysphoria, impairment, auditory alterations, and ego dissolution being more pronounced under psilocybin. Both compounds caused psychomotor slowing and spatial memory impairments, but did not produce empathogenic effects. The duration of self-reported effects of 2C-B was shorter than that of psilocybin, largely resolving within 6 hours. These findings support the classification of 2C-B as a moderately deep psychedelic. Tailored dose-effect studies are needed to understand the pharmacokinetic dependency of 2C-B's experiential effects.

Acute Effects of Psilocybin After Escitalopram or Placebo Pretreatment in a Randomized, Double-Blind, Placebo-Controlled, Crossover Study in Healthy Subjects

Anna M. Becker et al.

Summary: The study examined the interaction between escitalopram and psilocybin in the treatment of depression and anxiety. Escitalopram pretreatment was found to reduce certain negative effects of psilocybin, but further research is needed to fully understand the interactions between antidepressants and psilocybin in patients with psychiatric disorders.

Flashback phenomena after administration of LSD and psilocybin in controlled studies with healthy participants

Felix Mueller et al.

Summary: Reoccurring drug-like experiences after the administration of LSD and psilocybin are common in healthy subjects, occurring in up to 9.2% of participants. However, these experiences are generally mild and perceived as neutral to pleasant. Flashback phenomena do not appear to be clinically relevant in controlled studies with healthy participants.

Summary: Metabolomics analyzes metabolites in biological samples, particularly blood and its liquid components: plasma and serum, using advanced analytical chemistry methods. It has been found that small molecules play diverse roles in living systems beyond traditional building blocks or cellular energy fuel, leading to practical applications in molecular medicine and a better understanding of fundamental biochemical processes.

Metabolomics and integrated network analysis reveal roles of endocannabinoids and large neutral amino acid balance in the ayahuasca experience

Francisco Madrid-Gambin et al.

Summary: This study investigates the human metabolomics signature after consuming the psychedelic ayahuasca and its connection to subjective effects and plasma concentrations of ayahuasca alkaloids. The results suggest a crosstalk between circulating large-neutral amino acids and subjective effects, independent of alkaloid concentrations. Dysregulation in neurotransmission pathways such as serotonin and dopamine synthesis is also observed.

Summary: This study explores the analgesic effects of low doses of LSD without producing profound mind-altering effects. LSD at a low dose was found to increase pain tolerance and reduce subjective levels of experienced pain and unpleasantness. Further research into the analgesic effects of low doses of LSD in patient populations is warranted based on the findings.

Psychedelics in Psychiatry: Neuroplastic, Immunomodulatory, and Neurotransmitter Mechanisms

Antonio Inserra et al.

Summary: Psychedelic compounds are emerging as potential novel therapeutics in psychiatry, with their main effects mediated through the modulation of neurotransmitter systems, immunity, and neuroplasticity. Promising results suggest an opportunity for improvement in the treatment of psychiatric disorders, an area that has seen limited advancement in the past 20 years. Studies are underway to separate the psychedelic effects from the therapeutic effects of these compounds, aiming to provide more effective treatment options.

Psilocybin-induced changes in brain network integrity and segregation correlate with plasma psilocin level and psychedelic experience

Martin K. Madsen et al.

Summary: The novel therapeutic psilocybin exerts its psychedelic effects by engaging cerebral serotonergic targets with its active metabolite psilocin. The serotonin 2A receptor plays a critical role in altering neural processes, leading to increased entropy, reduced network integrity, and decreased segregation between brain networks. Psilocin is shown to shape the time course and magnitude of changes in cerebral functional architecture and subjective experience, providing novel insight into the neurobiological mechanisms underlying the psychedelic experience and consciousness.

Psychedelics and Neuroplasticity: A Systematic Review Unraveling the Biological Underpinnings of Psychedelics

Summary: Clinical studies suggest that psychedelics induce rapid changes in neuroplasticity mechanisms, leading to alterations in gene and protein expression, increased neurotrophic factor levels, and enhanced dendritic complexity. Repeated administration of psychedelics can directly stimulate neurogenesis and elevate BDNF mRNA levels, indicating potential long-lasting effects on brain plasticity. Future research could focus on deciphering specific cellular mechanisms activated by different psychedelics to better understand their therapeutic potential.
