Age-associated epigenetic changes in mammalian sperm: implications for offspring health and development

Summary: This article reviews age-related epigenetic changes in sperm in mammals and humans, including DNA methylation, histone modifications, and non-coding RNAs. Age affects all known epigenetic mechanisms in sperm, which may have implications for embryonic and offspring epigenetic and phenotypic events. Chemical and lifestyle exposures, as well as assisted reproductive technologies, can influence the epigenetic aging of sperm. The age-related changes in the sperm epigenome may compromise reproductive health of fathers and transfer altered epigenetic information to subsequent generations.

N-acetylcysteine rescues meiotic arrest during spermatogenesis in mice exposed to BDE-209

Summary: This study aimed to evaluate the protective effects of N-acetylcysteine (NAC) on meiotic arrest in spermatocytes and decreased sperm quality in BDE-209-exposed mice. The results showed that pretreatment with NAC attenuated the oxidative stress status induced by BDE-209, improved testicular histology and sperm quality, and enhanced DNA damage repair. Therefore, NAC may have a protective effect against the spermatogenesis dysfunction caused by BDE-209.

BDE-209 disrupted the blood-testis barrier integrity by inhibiting estrogen receptor α signaling pathway in Sprague-Dawley rats

Summary: This study aimed to investigate the role of estrogen receptor alpha (ER alpha) in BDE-209-mediated perturbation of BTB integrity. The data demonstrated that BDE-209 impaired BTB integrity by reducing crucial tight junction-related proteins and apical ectoplasmic specialization markers. Furthermore, BDE-209 damaged BTB ultrastructure including tight junctions and ectoplasmic specialization structures. Unexpectedly, PPT rescued BDE-209-mediated disruption of BTB integrity by activating ER alpha in Sertoli cells.

Declining semen quality and polybrominated diphenyl ethers (PBDEs): Review of the literature to support the derivation of a reference dose for a mixture risk assessment

Summary: By reviewing literature on polybrominated diphenyl ethers (PBDEs) and poor semen quality, this study finds associations between PBDE exposures and declining semen quality through epidemiological studies and rodent experiments. In vitro studies also show PBDE congeners acting as androgen receptor antagonists. Reference doses were estimated for specific PBDE congeners, which will be used in future mixture risk assessments for deteriorations in semen quality involving multiple chemicals.
