Early neurodevelopmental outcomes of extreme preterm infants exposed to paracetamol: a retrospective cohort study

Bella Zhong et al.

Summary: This retrospective cohort study investigated the effects of paracetamol exposure on early neurodevelopment in preterm infants. The study found no significant association between paracetamol exposure and early cerebral palsy or intellectual developmental abnormalities at 3-4 months corrected age.

Trial of paracetamol for premature newborns: five-year follow-up

Summary: The study indicates that using paracetamol in very preterm infants does not lead to common adverse effects. Most children showed normal development, with respiratory and allergic symptoms being less common, and neurologic issues being rare.

Long-Term Safety of Prenatal and Neonatal Exposure to Paracetamol: A Systematic Review

Ram Patel et al.

Summary: This study investigated the long-term safety of prenatal and neonatal paracetamol exposure. The available evidence suggests a possible association between prenatal paracetamol exposure and an increased risk of neurodevelopmental, atopic, and reproductive adverse outcomes.

Paracetamol (Acetaminophen) and its Effect on the Developing Mouse Brain

Gaetan Philippot et al.

Summary: This study evaluates the effects of neonatal exposure to clinically relevant doses of acetaminophen on adult behavior and brain development. The findings suggest that exposure to acetaminophen can lead to altered behavior and reduced cognitive abilities in adulthood, as well as oxidative stress in the hippocampus.

Intravenous paracetamol for neonates: long-term diseases not escalated during 5 years of follow-up

Sanna Juujarvi et al.

Summary: This study evaluated the long-term adverse reactions of paracetamol in children who required intensive care shortly after birth. The results showed that intravenous paracetamol given to neonates did not associate with childhood disorders compared with the non-exposed infants during the 5-year follow-up, thereby not confirming the hypothesis that early paracetamol use causes childhood morbidities.

Paracetamol (Acetaminophen) and the Developing Brain

Christoph Buehrer et al.

Summary: Paracetamol, commonly used during pregnancy for fever and pain, is suspected to increase the risk of attention deficit hyperactivity disorder and autism spectrum disorder in offspring by about 25% according to epidemiological studies. However, there are still unknown factors and mechanisms that require further investigation.

Prenatal and postnatal exposure to acetaminophen in relation to autism spectrum and attention-deficit and hyperactivity symptoms in childhood: Meta-analysis in six European population-based cohorts

Silvia Alemany et al.

Summary: The study found that children prenatally exposed to acetaminophen were more likely to have borderline or clinical ASC and ADHD symptoms in the future. The association was slightly stronger among boys, but postnatal exposure to acetaminophen did not show any significant association with ASC or ADHD symptoms. The results support the need to provide clear information to pregnant women and their partners about the potential risks of acetaminophen use.
